急性肺损伤小鼠肺泡巨噬细胞TREM1 mRNA的表达及意义 被引量:4

摘要 目的:研究脂多糖(LPS)致急性肺损伤(ALI)小鼠肺泡巨噬细胞(AM)髓系细胞触发受体1(TREM1)mRNA的表达及意义。方法:48只小鼠分成对照组和实验组,对照组腹腔注射生理盐水,实验组腹腔注射LPS诱导ALI模型;于灌注后3、6、12及24 h,采用荧光定量逆转录(RT-PCR)方法检测支气管肺泡灌洗液中提取的AM中TREM1 mRNA表达水平,用ELISA法检测支气管肺泡灌洗液上清液中的TNFα、可溶性TREM1(sTREM1)水平,并分析TREM1 mRNA与sTREM1和TNFα表达水平的相关性。结果:各对照组AM均表达TREM1 mRNA,但各时间点比较差异无统计学意义(P>0.05);观察组TREM1 mRNA、TNFα及sTREM1的表达均在3 h时上升致最高,之后下降;TREM1mRNA与sTREM1及TNFα的表达水平呈正相关(r=0.783,P<0.001;r=0.472,P<0.05)。结论:ALI小鼠支气管肺泡灌洗液、及其AM中TREM1 mRNA表达水平升高,TREM1 mRNA升高与sTREM1、TNFα呈正相关。 Objective: To investigate the expression and clinical significance of TREM1 mRNA in the alveolar macrophages( AM) of acute lung injury induced by lipopolysaccharide( LPS) in rat. Methods: 48 rats were randomly divided into control group and experiment group,control group was intraperitoneally injected with normal saline; experiment group was intraperitoneally injected with LPS to induce ALI model. After 3,6,12 and 24 h,RT-PCR was adopted to detect TREM1 mRNA expression level in AM extracted from bronchoalveolar lavage fluid. ELISA was used to detect the concentration of TNF-α and soluble TREM1 level in supernatant of bronchoalveolar lavage fluid; analyzing correlation between expression level of TREM1 mRNA and sTREM1 together with TNFα. Results: AM of control group expressed TREM1 mRNA,while comparison differences of each time point showed no statistical significance( P〉0. 05); the expression of TREM1 mRNA,TNFα and sTREM1 rose to the highest in 3 h,then declined; expression of TREM1 mRNA and TNFα and sTREM1 level were positively correlated( r = 0. 783,P〈0. 001 and r = 0. 472,P〈0. 05). Conclusion: ALI rat bronchoalveolar lavage fluid and expression level of TREM1 mRNA increased,the increase of TREM1 mRNA was positively correlated with sTREM1 and TNFα.
