舞茸菌丝体与胞外多糖液体培养基的优化

摘要 以舞茸(Grifola frondosa)LT菌株为材料,研究培养基营养成分对其菌丝体和胞外多糖的影响。通过单因素实验确定葡萄糖、蛋白胨、磷酸二氢钾及维生素B1为最适宜的碳源、氮源、无机盐和生长因子。多因素正交实验进一步优化上述组分的浓度,确定舞茸菌丝体最适宜的发酵培养基配方为:10%豆芽汁200 mL,葡萄糖20 g/L、蛋白胨5 g/L、磷酸二氢钾4 g/L、维生素B130 mg/L,pH 7.0;胞外多糖最适宜的发酵培养基配方为:10%豆芽汁200 mL,葡萄糖20 g/L、蛋白胨3 g/L、磷酸二氢钾4 g/L、维生素B130 mg/L,pH 7.0。研究为进一步开展舞茸菌丝体和胞外多糖液体发酵与利用提供了理论参考。 In the present study,a strain of Grifola frondosa LT was used to evaluate the effects of medium nutrition on the growth of mycelium and production of extracellular polysaccharides.Glucose,peptone,monopotassium phosphate and vitamin B1 were established as the optimal carbon source,nitrogen source,inorganic salt and growth factor via single factor test.The concentration of medium nutrition was further optimized through multi-factor orthogonal test,and the optimum fermentation condition for the growth of mycelium was 10% bean sprouts 200 mL,glucose 20 g/L,peptone 5 g/L,monopotassium phosphate 4 g/L and vitamin B1 30 mg/L,and the optimum fermentation condition for production of extracellular polysaccharides was 10%bean sprouts 200 mL,glucose 20 g/L,peptone 3 g/L,monopotassium phosphate 4 g/L and vitamin B130 mg/L.This research is of certain values to further fermentation researches and applications of G.frondosa mycelium and polysaccharides.
