Optimization in Microwave-Assisted Extraction Process of Total Flavonoids from Shaanxi Dianthus superbus by Response Surface Methodology

摘要 以陕西眉县的瞿麦为研究对象,以乙醇为提取剂,采用微波辅助提取工艺提取瞿麦中总黄酮。在单因素实验的基础上,以总黄酮得率为评价指标,采用响应曲面法确定最佳工艺为:料液比1∶20(g∶mL)、提取时间2min、乙醇体积分数60%,在此条件下,重复3次平行实验,总黄酮得率为1.113%,与模型预测值基本吻合,说明模型能较好预测陕产瞿麦中总黄酮得率,优化工艺条件较可靠。 Using Dianthus superbus from Mei county in Shaanxi as a research object, ethanol as an extractant, we extracted total flavonoids from Dianthus superbus by microwave-assisted extraction process. On the basis of the single-factor experiment, using the yield of total flavonoids as an index,we optimized the extraction conditions by a response surface methodology as follows.solid-liquid ratio was 1 : 20(g : mL), extraction time was 2 min,and volume fraction of ethanol was 60%. Under above conditions, the yield of total flavonoids for three parallel experiments was 1. 113 %, which approximately agreed with the model prediction value. Overall, the model can better predict the yield of total flavonoids from Shaanxi Dianthus superbus,and the optimized extraction process is more reliable.
