mmu-miR-3475-3P在心脏发育中的生物信息学分析

摘要 目的探讨mmu--miR-3475-3P可能参与的生物学过程及信号通路。方法用实时荧光定量PCR方法测定mmumiR-3475-3P在小鼠胚胎心脏和成熟心脏中的表达。再综合应用TargetScan、miRDB、miRanda等常用microRNA在线数据库对miR3475-3p进行靶基因预测,对所得靶基因进行基因功能注释(Go)和信号通路分析。结果 mmu-miR-3475-3P在小鼠胚胎心脏和成熟心脏中存在明显的表达差异,运用TargetScan、miRDB、miRanda对miR3475-3P进行生物信息学分析发现该microRNA可能调控441个靶基因。结论 mmu-miR-3475-3P在小鼠胚胎心脏高表达,其预测的靶基因富集于多个信号通路及细胞生物学过程。 Objective To investigate the mmu-miR-3475-3P possible participating in biology process and signal pathway. Methods The expression of mmu-miR-3475-3P in the mature mouse heart and in the embryonic mouse heart was measured by qRT-PCR. The target genes were predicted through comprehensively using the common microRNA on-line databases such as Tar- getScan, miRDB and miRanda, and then the obtained targeted genes were performed the gene function annotation and signal pathway analysis. Results There was significant difference between the expression of mmu-miR-3475-3P in the embryonic mouse heart and the expression in the mature heart. Bioinformatics analysis by using TargetScan,miRDB and MiRanda on miR3475-3P revealed that microRNA was likely to regulate 441 target genes. Conclusion mmu-miR-3475-3P is highly expressed in the embryonic mouse heart. The target genes predicted by mmu-miR-3475-3P are enriched in multiple signal pathways and cellular biological processes.
