Correlation between Long Noncoding RNA (lncRNA) and Radiotherapy Sensitivity in Multidrug Resistant Breast Cancer Patients

摘要 目的探究长非编码RNA与乳腺癌多药耐药患者放疗敏感性的相关性。方法 BS517、BS524、BS525-1590、SKBR-3、MCF-7共5种人乳腺癌细胞株,采用细胞梯度照光细胞克隆实验检测5种细胞株的存活分数(SF)。采用高通量lncRNA芯片筛选BS517、BS524、BS525-1590细胞株表达量较高的lncRNA,并分析lncRNA与放疗抵抗性的相关性。结果各细胞经梯度照光后,各细胞株的SF值大小排列(由大到小)依次为BS517、BS524、BS525-1590、SKBR-3、MCF-7,该顺序同样由高到低地反映出各细胞株对放疗的敏感性;R05532、NR-015441、NR-033374 3种lncRNA在细胞系中表达明显较高;R05532、NR-015441、NR-033374 3种lncRNA的表达水平与细胞放疗抵抗性呈正相关,其γ值分别为0.931、0.926、0.928,且P<0.05;AA745020的表达水平与细胞放疗抵抗性无明显的相关性,γ=0.416,P>0.05。结论R05532、NR-015441、NR-033374 3种lncRNA在人乳腺癌细胞中高表达,且当其高表达时呈现的放疗抵抗性最高,这3种lncRNA高表达时可作为放疗抵抗的预测分子。 Objective To study the correlation between long noncoding RNA( lncRNA) and radiotherapy sensitivity of multidrug resistance in breast cancer. Methods 5 kinds of human breast cancer cell lines of BS517,BS524,BS525-1590,SKBR-3 and MCF-7 were used to detect the survival fraction( SF) by cell gradient photographers. Long noncoding RNA( lncRNA) with high expression of BS517,BS524 and BS525-1590 were screened by high-throughput lncRNA chip,and the correlation between lncRNA and radiotherapy resistance was analyzed. Results After the cells were irradiated with gradient,the SF values of each cell line were in the order of BS517,BS524,BS525-1590,SKBR-3 and MCF-7,which was also reflected resistance to radio-therapy from high to low. The expression of 3 kinds of lncRNA likes R05532,NR-015441,NR-031374 in the cell lines was significantly higher than others. The expression levels of R05532,NR-015441 and NR-033374 were positively correlated with the radiotherapy resistance( γ = 0. 931,γ = 0. 926,γ = 0. 928),( P〈0. 05). There was no significant correlation between AA745020 expression level and cell radioactivity resistance,γ = 0. 416,P〉0. 05. Conclusion Three kinds of lncRNA such as R05532,NR-015441 and NR-033374 are highly expressed in human breast cancer cells and have the highest resistance to radiotherapy when they are highly expressed. These 3 genes can be used as predictors of radiation resistance.
