摘要
目的:观察Rab1A(Ras-related protein Rab-1A)对非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞系迁移的影响及其分子机制。方法:应用siRNA干扰Rab1A基因表达;应用RTCA(real-time cell analysis)法分析干扰Rab1A基因表达对NSCLC细胞迁移速率的影响;应用免疫印迹观察干扰Rab1A基因表达对JAK1(Janus Kinase 1)磷酸化水平的影响。结果:与阴性对照组相比,Rab1A siRNA转染组NSCLC细胞系的Rab1A蛋白表达水平显著下调(P<0.01);干扰Rab1A基因表达抑制NSCLC细胞迁移(P<0.01);干扰Rab1A基因表达可下调NSCLC细胞迁移相关蛋白JAK1磷酸化水平(P<0.01)。结论:Rab1A通过调节JAK1磷酸化水平影响NSCLC细胞迁移。
Objective :To investigate the effects of RablA (Ras -related protein Rab -1A) on the migraton of non - small cell lung cancer (NSCLC) cell hnes and its molecular mechanism. Methods :The siRNA interference was used to knockdown Rabl A. The effects of knocking Rabl A siRNA on the migraton of NSCLC cell lines were investiga ted by RTCA (real- time cell analysis). Western blot was used to analyze the effects of RablA on the phosphorylation level of JAK1 (P - JAK1 ). Results:Expression level of RablA protein was significantly down - regulated in Rabl A siRNA groups compared to negative control group (P 〈 0.01 ). Rabl A siRNA knockdown inhibited the migraton of NSCLC cell lines in RablA siRNA groups compared to negative control group ( P 〈 0.01 ). Phosphorylation ltev el of JAK1 (P- JAK1 ) was significantly decreased by RablA knockdown in RablA siRNA groups compared to nega- tive control group (P 〈 0.01 ). Conclusion: RablA promotes the migraton of NSCLC cell lines through regulating phosphorylation level of JAK1 ( P - JAK1 ).
