摘要
目的探讨不同浓度的Mn^(2+)处理对甘草不同器官活性成分分布差异的影响。方法以一年生甘草移栽苗为实验材料,通过盆栽蛭石的方法,共设置5个锰质量浓度水平,分别为0(对照)、1.81、18.1、36.2和54.3 mg/L,采用溶剂萃取法提取甘草中总黄酮,水提醇沉法提取甘草中总多糖,超声辅助法提取甘草中总皂苷,并利用紫外-可见分光光度法测定甘草不同器官3种成分含量差异。结果随着Mn^(2+)处理浓度的增加,甘草根中总皂苷含量较高,在1.81 mg/L浓度处理时达到最大值为17.28%,且各处理浓度间差异极显著(P<0.01);随着Mn^(2+)处理浓度的增加,甘草叶中总黄酮含量普遍较高,在54.3 mg/L浓度处理时达到最大值为6.38%,但是各处理浓度间差异不显著(P>0.05);随着Mn^(2+)处理浓度的增加,甘草根和茎中总多糖含量较高,且各处理浓度间存在显著性差异(P<0.05)。结论不同浓度的Mn^(2+)处理对甘草不同器官活性成分含量的分布有一定影响。
Objective To discuss the effects of different concentrations of Mn2+ on the contents of active components in different organs of Glycyrrhiza uralensis Fisch. Methods The transplants of one-year-old G. uralensis were subjected to five concentration of Mn(0, 1.81, 18.1, 36.2 and 54.3 mg/L) culturing in vermiculite and 1.81 mg/L Mn2+ was used as the normal concentration in the complete Hoagland nutrition solution. Using solvent extraction method for total flavonoids from G. uralensis, water extraction and alcohol precipitation method for polysaccharide, assisted method by ultrasonic wave extraction of total saponins, and the contents of three components in different organs of G. uralensis were determined by UV-visible spectrophotometric. Result With the increase of the concentration of Mn2+, the total saponin contents of roots of G. uralensis were higher than stems and leaves, and under 1.81 mg/L concentration treatment reached maximum value of 17.28%, and extremely significant differences in different concentrations(P 〈 0.01).With the increase of the concentration of Mn2+, the total flavonoids contents of leaves of G. uralensis were generally higher than roots and stems, and under 54.3 mg/L concentration treatment of Mn2+ reached maximum value of 6.38%, but no significant difference in different concentrations(P 〉 0.05). With the increase of the concentration of Mn2+, the total polysaccharides contents of roots and stems of G. uralensis were higher than leaves, and significantly differences in different concentrations(P 〈 0.05). Conclusion The treatments of different concentrations of Mn2+ could produce influnces on the contents of active components in different organs of G. uralensis.
