褐藻多糖硫酸酯、舒血宁注射液和丹红注射液对神经细胞保护的影响比较 被引量:1

摘要 目的探讨褐藻多糖硫酸酯、舒血宁注射液和丹红注射液对人神经母细胞瘤(SH-SY5Y)细胞增殖及缺糖缺氧保护的影响。方法在体外培养的SH-SY5Y细胞中加入不同浓度的褐藻多糖硫酸酯、舒血宁注射液和丹红注射液,处理不同时间,采用MTT法检测细胞活性;对SH-SY5Y细胞进行糖氧剥夺处理,加入不同浓度的三种药物处理细胞,分析三种药物对SH-SY5Y细胞增殖及缺氧缺糖导致的细胞损伤的影响;采用流式细胞术检测三种药物处理后细胞的周期变化。结果三种药物在一定浓度范围内促进细胞增殖。0.8 g/L褐藻多糖硫酸酯作用48 h后显著促进细胞增殖,细胞增殖率提高63.6%;对SH-SY5Y细胞的糖氧剥夺损伤有保护作用,细胞增殖率比对照提高13.5%;明显增加S期细胞比例,作用24 h由10.89%增加至18.18%。10%舒血宁注射液作用48 h后显著促进细胞增殖,细胞增殖率提高55.0%;对细胞糖氧剥夺损伤有保护作用,细胞增殖率比对照提高20.1%;对细胞周期无明显影响。5%丹红注射液作用48 h后显著促进细胞生长,细胞增殖率提高89.4%;明显增加S期细胞比例,作用72 h由15.05%增加至22.0%;但对细胞缺氧缺糖损伤无明显影响。结论一定浓度的褐藻多糖硫酸酯、舒血宁和丹红在体外均显著促进SH-SY5Y细胞的增殖,褐藻多糖硫酸酯和舒血宁降低缺氧缺糖导致的细胞损伤。三种药物均具有有效的神经细胞保护作用。 Objective To investigate the effect of Fucoidan, Shuxuening injection and Danhong injection on proliferation and protection of oxygen-glucose deprivation (OGD) of human SH-SY5Y neuroblastoma cells. Methods SH-SY5Y ceils were treated with different concentrations of Fucoidan, Shuxuening injection and Danhong injection for different time. Cell viability was determined by u- sing MTI" assay. SH-SY5Y cells were preconditioned for OGD. After treatment with the three types of drugs with different concentra- tions ,cell viability was detected in order to analyze the effect of the drugs on cell proliferation and injure induced by OGD. Flow cytome- try was used to analyze the cell cycle changes. Results The three types of drugs at certain concentration increased the cell prolifera- tion. O. 8 g/L of Fucoidan treatment for 48 h,a 63.6% increase in the cell growth was observed. For protection of cell injury caused by OGD,the proliferation rate was increased by 13.5% and percentage of S phase was markedly increased from I0. 89% to 18. 18% after treatment for 24 h when compared to the controls. 10% Shuxuening injection treatment for 48 h, a 55.5% increase in cell growth was found. For protection of cell injury caused by OGD, the proliferation rate was increased by 20. 1% but no obvious effect on the cell cycle distribution was observed when compared to the controls. 5% Danhong injection treatment for 48 h, a 89.4% increase in the cell growth and a significantly increase in the cell percentage of S phase from 15.05% to 22. 0% after 72-hour treatment were observed,but no ob- vious effect on OGD injury was found. Conclusion Fucoidan, Shuxuening injection and Danhong injection at certain concentration sig- nificantly increase the proliferation of SH-SY5Y ceils. Fucoidan and Shuxuening injection could reduce the cell injury caused by OGD. The three types of drugs all have a protective effect on neural cells.
