摘要
目的考察金花茶叶不同极性提取物对胰岛素抵抗HepG2细胞葡萄糖消耗量的影响及对Ⅱ型糖尿病小鼠的降血糖作用。方法利用胰岛素诱导HepG2细胞建立胰岛素抵抗模型,根据细胞对葡萄糖消耗情况,评价金花茶降糖活性。利用高脂高糖饲料联合链脲佐菌素建立小鼠Ⅱ型糖尿病模型,连续灌胃28 d观察血糖变化及糖耐量水平,同时检测血清总胆固醇、甘油三酯及低密度脂蛋白含量。结果金花茶叶正丁醇和乙酸乙酯提取物均能显著增加胰岛素抵抗HepG2细胞的葡萄糖消耗量(P<0.05)。并且在给药28 d后均能显著降低糖尿病小鼠血糖值和餐后血糖(P<0.01),同时显著降低糖尿病小鼠的总胆固醇含量、甘油三酯含量及低密度脂蛋白含量(P<0.05)。结论金花茶叶正丁醇和乙酸乙酯提取物能改善HepG2细胞的胰岛素抵抗现象,且能改善Ⅱ型糖尿病小鼠的高血糖高血脂症状。
Objective To investigate the effect of different polarity fractions of Camellia chrysantha (Hu) Tuyama leaves on glucose consumption in insulin resistance HepG2 cells as well as the hypoglycemic effect in type II diabetic mice. Methods High active hypoglycemic fraction was determined by glucose consumption improvement in insulin resistance HepG2 cell line induced by coculture with insulin in vitro. High fat and high sugar diet combined with streptozotocin induced type II diabetes mice model. The blood glucose levels, glucose tolerance as well as the total serum cholesterol, triglycerides and low density lipoprotein levels were detected after 28 days intragastric administration. Results The n-butanol fraction and ethyl acetate fraction significantly increased glucose consumption in insulin resisted HepG2 cell (P 〈 0.05). And both fractions significantly reduced blood glucose and postprandial serum glucose (P 〈 0.01), as well as reduced the level of total cholesterol, triglyceride content and low density lipoprotein significantly (P 〈 0.05) in diabetic mice after 28 days treatment. Conclusion The n-butanol and ethyl acetate fraction of Camellia chrysantha (Hu) Tuyama can improve the insulin resistance phenomenon of HepG2 cell also alleviate the symptoms of hyperglycemia and hyperlipidemia in type II diabetic mice.
