摘要
目的探讨2型单纯疱疹病毒(HSV-2)感染后人角质形成细胞分泌细胞因子的状况。方法采用非洲绿猴肾细胞株VERO细胞进行HSV-2病毒扩增,再以HSV-2感染Ha Ca T细胞,采用ELISA和荧光实时定量PCR检测感染后24h、48h和72h时IL-10、IFN-γ、TGF-β的分泌和表达情况。结果经HSV-2感染后Ha Ca T细胞分泌和表达的IL-10、IFN-γ、TGF-β与对照组相比有统计学差异,并呈时间相关性。结论在HSV-2感染早期KC参与了免疫反应,其分泌和表达的细胞因子的偏移可能参与了HSV-2的病理机制。
Objective To probe the state of cytokine level in the HSV-2-infected human keratinocyte(KC).Methods VERO cells were used for the propagation and titration of the HSV-2, and HaCaT ceils were infected with HSV-2,then were harvested 24h, 48h and 72h.Expression levels of IL-10,IFN-Y and TGF-13were detected by ELISA and Real-time PCR.Results We infected HaCaT cells with HSV-2,1evels of IL-10,IFN-Y and TGF-βchanged significantly in a time-dependent manner (P〈0.05).Conclusion Ker- atinocytes participate in the immune response on the early stage of HSV-2 infection, the shift of antiviral cytokine may involved in this process.
