ILK及E-cadherin在结肠癌中的表达及其预后意义

Prognostic significance and expression of ILK and E-cadherin in colon carcinoma

[目的]探讨整合素连接激酶(integrin-linked kinase,ILK)和E-钙黏蛋白(E-cadherin)在结肠癌发生中的意义。[方法]采用免疫组织化学S-P法检测ILK和E-cadherin在50例结肠恶性肿瘤、25例正常结肠标本中的表达情况,采用Western blot法检测ILK和E-cadherin在30例结肠恶性肿瘤及正常结肠标本中的表达。[结果]免疫组化结果:结肠癌组织中ILK阳性表达率为66.0%(33/50)明显高于正常结肠组织24.0%(6/25);而E-cadherin阳性表达率为42.0%(21/50)则明显低于正常结肠组织中的表达96.0%(24/25);ILK与E-cadherin在结肠癌中的异常表达与浸润深度和淋巴结转移有关(P<0.05)。Western blot结果显示:ILK在结肠癌组织中的表达显著高于正常组织(P<0.05)。[结论]ILK及E-cadherin与结肠癌侵袭、转移行为密切相关,ILK和E-cadherin的联合检测将有助于判断结肠癌患者的预后。

[Objective]To study the clinical significance of integrin-linked kinase（ILK）and E-cadherin in colon carcinoma.[Methods]The expression of ILK and E-cadherin proteins were detected by immunohistochemical staining（Streptavidine-Peroxidase method）in 50 cases of colon carcinoma and 25 cases of adjacent normal colon.The ILK and E-cadherin protein level were detected by Western blot method in 30 cases of colon carcinoma and adjacent normal colon.[Results]The positive rate of ILK was significantly higher in the colon carcinoma group 66.0%（33/50）than in the adjacent normal colon 24.0%（6/25）.However,lower positive rate of E-cadherin was detected in colon carcinoma group 42.0%（21/50）than that in adjacent normal colon group 96.0%（24/25）.The abnormal expression of ILK and E-cadherin were significantly correlated with the depth of invasion and lymph node metastasis（P〈0.05）.Western blot showed that the level of ILK in the colon carcinoma was higher than that in the adjacent normal colon（P〈0.05）,while the level of E-cadherin inthe colon carcinoma was lower than in the adjacent normal colon（P〈0.05）.[Conclusion]The expression of ILK and E-cadherin is significantly correlated with tumor invasion and metastasis.The detections of ILK and E-cadherin will help us to predict prognosis of patients.
