叶黄素、欧洲越橘提取物联合使用大鼠30d毒性观察

Observation on the toxicity in rats by using the extract of lutein and bilberry jointly for 30 days

摘要 目的了解叶黄素、欧洲越橘提取物联合使用对大鼠30 d毒性表现。方法每天按0.417、0.833、1.667 g/(kg·BW)的叶黄素、欧洲越橘提取物混合物给予大鼠灌胃30 d,记录体重和食物利用率,实验结束时采血作血生化及血常规检测,统计脏器系数并作肝、肾、脾、胃、睾丸、卵巢病理学检查。结果叶黄素、欧洲越橘提取物联合使用对大鼠生长发育无不良影响(雄性体重F=0.965、进食量F=3.828、食物利用率F=0.062,雌性体重F=0.813、进食量F=3.535、食物利用率F=0.1 462),血液学、血清生化指标值仅雄性中剂量组嗜酸性粒细胞(F=1.885),雌性高剂量组嗜碱性粒细胞(F=2.193)、雄性高剂量组肌酐(F=2.773)与对照组比较,差异有统计学意义(P<0.05),但均在正常值范围内。主要脏器外观及脏器系数(雄性肝体比F=0.865、肾体比F=0.382、脾体比F=0.606、睾体比F=0.321;雌性肝体比F=2.283、肾体比F=0.599、脾体比F=0.198)未见异常,组织病理学检查未发现特异性病变。结论叶黄素、欧洲越橘提取物联合使用在30 d周期内未见大鼠有明显毒性表现。 Objective To understand the 30 days toxicity affect by using the extract of lutein and bilberry jointly. Methods In 30 days,the rats were given by gavage the combination of extract of lutein and bilberry at doses of 0. 417,0. 833 and 1. 667 g/( kg·BW) every day. The weight of rats and food conversion efficiency were recorded. At the end of the test,the blood of rats was drawn to detect the blood routine and blood biochemical indexes. Organ coefficient was calculated. The pathological examination was made on liver,kidney,spleen,stomach,testicle and ovary. Results The use of the extract of lutein and bilberry jointly had no unpleasant effects on the growth of rats( Male rats: weight F = 0. 965,food intake F = 3. 828,food conversion efficiency F = 0. 062. Female rats: weight F = 0. 813,food intake F = 3. 535,food conversion efficiency F = 0. 1 462). For blood routine and blood biochemical indexes,compared with control group,only eosinophilic granulocyte in medium dose group of male rats( F = 1. 885),basophilic granulocyte in high dose group of female rats( F = 2. 193),and creatinine in high dose group of female rats( F = 2. 773) had significant differences( P 〈 0. 05). However,all differences were within the normal range. There was no difference for the appearance of main organs and organ coefficients( Male rats: liver/body ratio F = 0. 865,kidney/body ratio F = 0. 382,spleen/body ratio F = 0. 606,testicle/body ratio F = 0. 321. Female rats; liver/body ratio F = 2. 283,kidney/body ratio F= 0. 599,spleen/body ratio F = 0. 198). Histopathologic examination showed no specific lesion. Conclusion No obvious toxicity was found in rats by using the extract of lutein and bilberry jointly for 30 day.
