摘要
本研究以甜瓜杂交新品种‘西州密25号’和‘西州密17号’及其亲本种质为试验材料,利用SSR(simple sequence repeats)分子标记检测技术,并结合大田传统鉴定方法对其种子纯度及真实性进行了分析研究。结果表明,从18对SSR引物中筛选得到多对特异性引物可以鉴定出‘西州密25号’与‘西州密17号’杂交种的纯度。‘西州密25号’三批种子纯度分别为98.42%、99.41%、99.38%;‘西州密17号’的种子纯度为99.22%。SSR分子标记鉴定结果与‘西州密25号’和‘西州密17号’的大田植物学形态特征鉴定结果符合率高达99%以上。SSR分子标记方法可以准确鉴定出杂交种纯度及真实性,且大大缩短鉴定周期,可为开展‘西州密25号’和‘西州密17号’甜瓜新品种的杂交种纯度室内鉴定快速检测提供技术支撑。
In this research, simple sequence repeat(SSR) molecular markers were used to test variety purity and authenticity identification of new melon hybrids‘Xizhoumi 25'and‘Xizhoumi 17', and we used them and their parents as experimental materials, and the results were verified by morphological identification. Several pairs of specific primers out of 18 pairs of SSR core primers were selected to make a rapid variety purity test. The seed purity of 1, 2, 3 batch of ‘Xizhoumi 25'and ‘Xizhoumi 17'detected by SSR markers were 98.42%, 99.41%,99.38%, and 99.22%,respectively. Compared with results of morphological identification of ‘Xizhoumi 25'and‘Xizhoumi 17'in field, the coincidence rate of the variety purity detected by SSR markers were higher than 99%.SSR molecular marker method could identify hybrid purity and authenticity accurately, and shorten the appraisal period greatly. The results showed that the technical system can meet the requirements of rapid variety purity test of the melon new hybrids‘Xizhoumi 25'and‘Xizhoumi 17'in laboratory.
