摘要 本研究采用水平板琼脂糖凝胶和垂直板聚丙烯酰胺凝胶分别对微卫星PCR产物进行电泳分离,利用硝酸银染色法和荧光染料染色法染色,从染色背景、染色时间及分辨率角度对不同凝胶电泳分离及染色方法进行比较,探讨微卫星PCR两种凝胶电泳及不同染色方法的特点及适用性。结果显示,常规银染法较快,染色液可重复利用,是一种经济实用的染色方法,但操作繁琐,染色液含有甲醛,具有一定毒性。无毒的GelRed荧光染料染色速度快,操作简单,是一种比较成功的凝胶电泳染色法。 In the study, microsatellite PCR products were separated by horizontal plate agarose gel and vertical plate polyacrylamide gel, respectively. The samples were stained by the methods of nitrate silver staining and fluorescent dye staining. Different gel electrophoresis separation and staining methods were compared from the dyeing background, dyeing time and resolution, which to explored two kinds of gel electrophoresis and different staining methods of the characteristics and applicability in the microsatellite PCR. The results showed that the conventional silver staining method was rapid and the dyeing solution can be reused. It was a practical dyeing method, but the operation was complicated, and the dyeing liquid contained formaldehyde, which had certain toxicity. Non-toxic Gel Red fluorescent dye staining was a more successful gel electrophoresis staining which had high speed and simple operation.
