摘要
为了研究丝瓜超氧化物歧化酶(superoxidedismutase,SOD)酶学特性,本研究应用分光光度计开展了温度、pH、酶体积、反应底物浓度等对酶反应的影响,为丝瓜酶促褐变的研究提供基础。试验结果表明SOD酶最优反应体系为:以底物氮蓝四唑(NBT)为例,最优温度为40℃,最适pH为7.8,其底物浓度为0.08 mol/L,最适酶液体积为0.14 m L,其根据米氏方程:y=0.002 7x-0.013(x=1/V,y=1/S),Km为0.207 9 mol/L。
The factors influencing the enzymatic characteristics of superoxide dismutase(SOD) in loofah were studied by spectrophotometry, which provided the basis for the study of enzymatic browning of loofah. Use the common loofah pulp as the material. In order to study the effects of reaction temperature, p H and measurement wavelength, substrate concentration and SOD activity of luffa. SOD optimum reaction system: NBT as substrate with optimum concentration 0.08 mol/L, optimum temperature 40℃, optimum p H 7.8, optimum enzymatic liquid volume 0.14 m L, and the Michaelis Menten equation is y=0.002 7x-0.013(x=1/V, y=1/S), Km 0.207 9 mol/L.
