晚期氧化蛋白产物致胆固醇敏感器SCAP功能失调导致巨噬细胞胆固醇含量增加

摘要 目的:探讨晚期氧化蛋白产物对巨噬细胞内胆固醇含量及SCAP表达的影响。方法:采用伏波醇酯(PMA)诱导THP-1单核细胞形成巨噬细胞,AOPP体外干预THP-1源性巨噬细胞,油红O染色鉴定,应用酶法测定巨噬细胞内胆固醇含量,应用Real-time PCR与Western blot方法测定巨噬细胞LDLr、SREBPs、SCAP mRNA及蛋白表达。结果:AOPP组巨噬细胞内胆固醇含量明显高于对照组,AOPP组巨噬细胞LDLr、SREBPs、SCAP mRNA及蛋白表达显著高于对照组。结论:晚期氧化蛋白产物能够增加巨噬细胞内胆固醇含量,SCAP功能障碍是细胞内胆固醇含量增加的主要原因之一。 Objective: To investigate the effect of AOPP in the macrophage cholesterol content and the impact of SCAP expression. Methods: Using 12-o- tetradecanoyl phobol- 13-acetate (PMA) induced THP- 1 monocytes to form macrophages, in vitro, AOPP were treated with THP - 1 - derived macrophages and identified by oil red O staining, The cholesterol content of macrophages was measured by enzymatic method. Real - time PCR and Western blot were used to detect the expression of LDLr, SREBPs, SCAP mRNA and protein in macrophages. Results: Macrophage cholesterol content in AOPP group was obviously higher than that of control group, the expression of LDLr, SREBPs, SCAP mRNA and protein of the group AOPP macrophages is significantly higher than that of the control group. Conclusion: Advanced oxidation protein products increases the cholesterol content of macrophage, intracellular cholesterol increases associating with cholesterol sensor SCAP dysfunction.
