摘要
目的侵袭与转移是影响膀胱癌患者疗效及导致死亡的主要因素,其具体机制尚不明确;研究表明miR-451与多种肿瘤的发生、发展密切相关。本研究分析miR-451在膀胱癌中的表达及其临床意义,观察miR-451表达改变对上皮型钙黏附蛋白(E-cadherin)和间质型骨架蛋白(Vimentin)表达的影响。方法收集2012-01-01-2015-12-31江西省人民医院收治的47例膀胱癌患者的癌组织及10例正常膀胱组织标本,运用实时荧光定量PCR(quantitative Real-time PCR,qPCR)检测miR-451的表达及运用免疫组织化学技术检测E-cadherin和Vimentin的表达,分析miR-451表达与膀胱癌临床病理因素,以及E-cadherin和Vimentin表达的关系。利用脂质体将miR-451拟似物(miR-451mimics)转染入膀胱癌5637细胞,采用蛋白质印迹法检测E-cadherin和Vimentin的表达。结果 miR-451在膀胱癌组织中的表达为0.14±0.017,明显低于正常膀胱组织(1.0),其表达和膀胱癌临床分期及病理分级密切相关,P<0.01;E-cadherin在膀胱癌组织较正常组织中低表达,分别为36.17%和84.50%;Vimentin在膀胱癌组织及正常膀胱黏膜层中表达不明显。癌组织中miR-451表达与E-cadherin表达正相关,r=0.67,P=0.023;miR-451过表达后,E-cadherin蛋白表达上调(P<0.001),Vimentin蛋白表达无明显改变,P>0.05。结论MiR-451是膀胱癌潜在的调控子,可能是通过调控E-cadherin蛋白表达进而影响膀胱癌发生及进展。
OBJECTIVE The mechanisms of bladder cancer cell metastasis and invasion mainly leading to unfavor- able treatment or death are not clear at present. Research have confirmed that miR-451 was closely related to tumorigene- sis in most tumors. The aim of this study was to explore the expression and clinical significance of miR-451 and its effect on the expression of E-cadherin and Vimentin in bladder carcinoma. METHODS Totally 47 bladder cancer tissues and 10 normal bladder tissues from the patients of bladder tumor following radial cystectomy in our hospital from 2012-01-01 to 2015-12-31 were collected. The expression of miR-451 in these tissues was detected by qPCR,the expression of E-cadherin and Vimentin was detected by immunohistochemistry. The relationship between expression of miR-451 and clinical patho- logical feature and expression of E cadherin and Vimentin was statistically analyzed. MiR-451 mimics were transfected into 5637 cells by LipofectamineTM 2000, E-cadherin and Vimentin protein was measured by Western-blot. RESULTS The miR-451 expression level was significantly lower in tumor tissues (0.14± 0. 017) than that in normal tissues (1. 0), P〈0. 001. The tumor grade and clinical stage was significantly correlated with miR-451 expression (P〈0.01). The E-cadherin expression level was significantly lower in tumor tissues (36.17%) than that in normal tissues (84.5%),but the Vimentin in the two groups both expressed unobviously. MiR-451 expression was positively correlated to E-cadherin ex- pression (r=0.67,P=0. 023). The E-cadherin expression in 5637 cells was up-regulated after transfected with miR-451 mimics (P〈0. 001) ,but the Vimentin expression didn't changed(P=0. 058). CONCLUSION MiR-451 is a potential reg- ulator of bladder carcinoma, it may influences the pathogenesis and development of bladder carcinoma by regulating the expression of E-cadherin.
