响应面分析法优化金线莲黄酮提取工艺的研究 被引量:6

Optimization of Flavonoids Extraction from Anoectochilus Roxburghii by Response Surface Methodology

摘要 为优化金线莲黄酮提取工艺,根据单因素试验结果与响应面分析法中的Box-Behnken中心组合设计原则选取试验因素与水平,从中选取对叶绿素提取结果有影响的提取时间、提取温度、乙醇体积分数、液料比4个因素进行优化;并利用Design Expert 7.1.6分析软件对试验数据进行分析。结果表明,影响金线莲总黄酮含量的因素大小顺序为:提取温度、乙醇体积分数、提取时间、液料比,金线莲黄酮提取的最佳工艺参数为提取温度62.25℃,乙醇浓度为80.68%,提取时间89.8 min,液料比为79.38∶1;在此条件下,金线莲黄酮含量为2.243 7 mg/g,与预测值2.338 6 mg/g接近。采用响应面分析法得到的金线莲黄酮提取优化工艺准确可靠、可行,可用于金线莲总黄酮的提取,为金线莲资源的进一步开发利用提供了实验依据。 Response surface methodology( RSM) was employed to optimize the extraction conditions of total flavonoids from Anoectochilus roxburghii. Based on single-factor tests,Box-Benhnken center composite experiment was carried out with four factors,including alcohol concentration,extraction temperature,extraction time and ratio of liquor to material. Meanwhile,the experimental data were analyzed by Design Expert 7. 1. 6 software. The results showed that the factors affecting the flaronoids content from Anoectochilus roxburghii were extraction temperature,alcohol concentration,extraction time and ratio of liquor to material. The optimum extraction conditions by RSM were extraction time 89. 8 min,extraction temperature 62. 25 ℃,alcohol concentration 80. 68% and ratio of liquor to material 79. 38∶ 1. Under these conditions,the extraction yield of total flavonoids was 2. 243 7 mg/g,which was close to the predicted value 2. 338 6 mg/g. Using the method of response surface methodology to analyze the optimization process of flavonoids extraction from Anoectochilus roxburghii is accurate,reliable and feasible,which is useful for the extraction of flavonoids from Anoectochilus roxburghii and provides the certain experimental basis for deep research of Anoectochilus roxburghii resources.
