含硒76肽对小鼠血糖的影响及其抗氧化作用 被引量:2

摘要 目的探索含硒76肽(Se-76P)皮下注射后不同时间对小鼠血糖(BG)的影响及其在肝中发挥的抗氧化作用。方法按不同剂量Se-76P(20μmol/kg、40μmol/kg、60μmol/kg、80μmol/kg、100μmol/kg)给予C57BL/6J小鼠皮下注射,以亚硒酸钠(40μmol/kg)和胰岛素(2.5 IU/kg)为对照检测注射后不同时间(30、60、90、120、150 min)BG的变化,同时测定肝组织中谷胱甘肽过氧化物酶(GPX)、超氧化物歧化酶(SOD)和脂质过氧化产物丙二醛(MDA)的含量变化。结果皮下注射较低剂量的Se-76P(20μmol/kg、40μmol/kg、60μmol/kg)可使小鼠BG在150 min内维持在较低水平,但效果低于胰岛素;较高剂量的Se-76P(80μmol/kg、100μmol/kg)可使小鼠BG在120 min内维持在较低水平,在150 min时恢复或略高于正常水平;而亚硒酸钠仅能在30 min内降低小鼠BG,然后促使BG升高。另外,Se-76P还可明显提高肝组织中GPX和SOD活力,抑制脂质过氧化产物MDA的产生。结论适量的Se-76P可在小鼠体内稳定地发挥降低BG的作用,并维持肝组织的氧化还原平衡。 Objective To explore the effect of subcutaneous injection of selenium 76 peptide(Se-76P) on blood glucose(BG) level and its anti-oxidation inmice at different times. Methods C57BL/6J mice were injected subcutaneously with different doses of Se-76P ( 20 μmol/kg,40 μmol/kg,60μmol/kg, 80μmol/kg, 100 μmol/kg), then the changes of BG level were detected at different time (30,60,90,120,150 min),while sodium selenite (Na2SeO3 ) (40 μmol/kg) and insulin (2.5 IU/kg) as the controls. Meanwhile, the activities of glutathione peroxidase (GPX), superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in mouse liver tissue were measured. Fiesuits The BG was maintained at a low level within 150 rain when the mice were subcutaneously injected with a lower dose of Se-76P (20 μmol/kg,40 μmol/kg,60 μmol/kg) ,but the effect is lower than insulin. Higher doses of Se-76P (80 μmol/kg, 100 μmol/kg) allowed the BG to remain at a low level within 120 rain and recovered or slightly above normal at 150 rain. While Na2SeO3 only reduced BG within 30 min and then increased BG in mice. In addition,Se- 76P also can significantly improvethe GPX and SOD activity, and obviously inhibit the production of hpid peroxidation product MDA in mouse liver tissue. Conclusion Appropriate Se-76P can not only play a role in reducing the BG level in mice, but also to maintain redox balance in the mouse liver tissue.
