摘要
目的:通过睡眠剥夺大鼠模型脑电图α波、β波的频率,分析针刺百会穴对失眠的治疗效果。方法:采用水平台法建立睡眠剥夺大鼠模型,分为实验组、假穴位组和模型组,以正常喂养的大鼠作为空白对照组。实验组行百会穴针刺治疗,假穴位组行百会穴左侧旁0.2cm的假穴位针刺治疗,模型组、空白组不予以治疗。在72h睡眠剥夺结束后记录各组大鼠脑电波数据,观察针刺百会穴对睡眠剥夺大鼠模型脑电图α波、β波频率的影响。结果:针刺前,实验组、假穴位组、模型组大鼠α波、β波频率无统计学差异(P>0.05),且α波均低于空白组,β波均高于空白组,说明睡眠剥夺可影响正常脑电波频率,提升大鼠大脑皮层兴奋性;针刺假穴位组大鼠脑电图变化与模型组比较无统计学差异(P>0.05),且治疗后大鼠的狂躁表现无显著改善,没有立刻进入睡眠;针刺治疗中,实验组大鼠α波频率上升、β波频率下降,趋向于正常,且治疗后立刻进入睡眠状态,说明针刺百会穴可降低睡眠剥夺模型的大脑皮层兴奋度。结论:针刺百会穴对睡眠剥夺大鼠模型具有安神作用。
Objective:To analyze the efficacy of acupuncturing at Baihui (DU 20) acupoint in the treatment for insomnia via monitoring the frequencies of α-wave,β-wave of EEG of rats deprived of sleep, Methods: The process of rat sleep was interrupted with platform-over-water protocol. Rats were randomized into 4 groups as sleep deprivation(SD; untreated), SD+ treatment (receive acupuncture at DU 20 acupoint), sham acupuncture (point selected at 0.2 cm aside left DU 20 acupoint), and control (normally fed and untreated). All EEG data were attained at 72nd hour as the end of sleep deprivation. Frequencies of α-wave and β-wave were analyzed for evaluating the influences of acupuncturing at DU 20 on the sleepless rats. Results : No differences were observed on the frequencies of α-wave and β-wave among groups of SD+ treatment, sham acupuncture and SD. The three groups showed lower α-wave frequency and higher β-wave frequency than the control, indicating that normal EEG frequency was influenced under sleep deprivation, which increased cerebral cortical excitability of the sleepless rats. Sham acupuncture and SD groups illustrated insignificant difference both in EEG and manifestation, and, no obvious improvement was observed on the rats' mania and was slow in falling asleep. SD rats that received acupuncture at DU 20 demonstrated increase in α-wave frequency and decline in β-wave frequency as well as falling asleep fast. This phenomenon implies that aeupuncturing at DU 20 lowers the cerebral cortical excitability of the rats suffered from sleep deprivation. Conclusion : Acupuncturing at DU 20 tranquilizes sleep deprived rat model.
