摘要
目的探讨茴香提取液对糖尿病大鼠肾脏损害的保护作用。方法按血糖分层随机将40只大鼠分为正常对照组(n=8)和造模组(n=32),给予造模组大鼠腹腔注射链脲佐菌素制备糖尿病模型,同时给予正常对照组腹腔注射等体积柠檬酸缓冲液。成模后,将造模组按随机数表法进一步分为茴香高剂量组、茴香低剂量组、二甲双胍组、模型组,每组各8只。每组给予相应处理30 d后,比较各组大鼠FBG、血清尿素氮(BUN)、肌酐(SCr),肾脏组织谷胱甘肽(GSH)、丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性,并分析苏木精-伊红染色下各组大鼠肾脏组织形态学改变。结果与正常对照组比较,模型组FBG、BUN、SCr及MDA水平均较高,而SOD和GSH水平较低(均P<0.05);与模型组比较,二甲双胍组、茴香组的FBG水平均较低;茴香高剂量组SCr、BUN水平均较低、肾组织SOD和GSH水平均较高,而茴香低剂量组仅SCr水平较低(均P<0.05)。茴香组肾小管的变性、坏死较模型组轻且管型少,茴香高剂量组比茴香低剂量组病变轻且范围小。结论一定浓度范围内的茴香提取液具有减轻糖尿病大鼠肾脏损伤、改善肾功能、提高肾组织抗氧化的作用。
modeling group was given intraperitoneal injection with streptozotocin to develop the diabetic rat model ,while the normal control group was given intraperitoneal injection of equivoluminal citrate buffer solution .After successful modeling ,the modeling group was further divided into high-dose Foeniculum vulgare group,low-dose Foeniculum vulgare group,metformin group and model group according to the random number table,with 8 cases in each group.After 30 days of the corresponding treatment ,FBG,blood urea nitrogen(BUN),serum creatinin (SCr),levels of L-glutathione(GSH) and malondialdehyde (MDA) in renal tissues,and activity of super oxide dismutase (SOD) were compared among groups,and morphological changes in kidney tissues using hematoxylin -eosin stain were analyzed among the rats in each group.Results Compared with the normal control group ,the levels of FBG,BUN,SCr and MDA were higher but the SOD and GSH levels were lower in the model group(all P〈0.05).Compared with the model group,the FBG level was lower in the metformin group and Foeniculum vulgare groups,the SCr and BUN levels were lower and the levels of SOD and GSH in kidney tissues were higher in the high-dose Foeniculum vulgare group,but the low-dose Foeniculum vulgare group had a lower SCr level(all P〈0.05).The Foeniculum vulgare groups obtained milder degeneration and necrosis as well as less tube type compared with the model group,and the high-dose Foeniculum vulgare group had milder lesions and smaller lesion range compared with the low-dose Foeniculum vulgare group.Conclusion Foeniculum vulgare extracts within a certain limits of concentrations might play a role in reducing the damages to the kidney of diabetic rats and improving renal function and kidney antioxidant effects .
