芪榔合剂对功能性便秘小鼠结肠AQP4和AQP8含量的影响研究 被引量:5

摘要 目的观察芪榔合剂对功能性便秘模型小鼠结肠组织中水通道蛋白4(AQP4)和水通道蛋白8(AQP8)表达的影响,探讨芪榔合剂治疗功能性便秘的作用机制。方法将40只健康昆明种小鼠按性别、体质量随机分为阴性对照组、便秘组、大剂量中药组、小剂量中药组,每组10只。阴性对照组给予50 mg/kg蒸馏水灌胃,其余组均予以50 mg/kg复方地芬诺酯灌胃20 d制作便秘模型。造模成功后第2天,阴性对照组、便秘组给予蒸馏水0.002mL/g灌胃7 d;大剂量中药组、小剂量中药组给予0.025 mL/g相应量中药(分别含生药2.9 g/mL和1.45 g/mL,相当于成人体质量30倍和15倍)灌胃7d。采用免疫组化法检测各组小鼠结肠组织中AQP4、AQP8表达情况。结果便秘组小鼠结肠组织中AQP4、AQP8含量均明显高于阴性对照组(P均<0.05);大剂量中药组、小剂量中药组小鼠结肠组织中AQP4、AQP8含量均明显低于便秘组,且小剂量中药组AQP8含量明显低于大剂量中药组(P<0.05)。结论便秘小鼠结肠组织中AQP4、AQP8含量明显升高,芪榔合剂对功能性便秘小鼠结肠组织中AQP4、AQP8含量具有一定的调节作用。 Objective It is to observe the effect of QiLang-HeJi on the expression of Aquaporin-4 (AQP4)and Aquaporin- 8 (AQP8) in colon tissue of mice with chronic constipation,and to study its treatment mechanism. Methods 40 healthy Kunming mice were randomly divided into control group, constipation group, high dose TCM group and low dose TCM groups, each group had 10 mice. The mice in the control group were given distilled water 50 mg/kg by lavage, the other mice were given Compound Diphenoxylate 50 mg/kg for establishing model of constipation. On the second day after model establishing successfully, the mice in control group and constipation group were given 0. 002 mL/g distilled water by lavage, the mice in high and low dose TCM group were respectively treated with 0. 025 mL/g Chinese medicine ( containing crude drug 2.9 g/mL and 1.15 g/mL respectively) for 7 days. The expression of AQP4 and AQP8 in colon tissue were detected by immunohisto- chemistry. Results The contents of AQP4 and AQP8 in colon tissue in constipation group was significantly higher than that in control group (P 〈 0.05 ) , and the expression in high and low dose TCM group was lower than that in constipation group ( P 〈 0.05), the expression of AQP8 in the low dose group was lower than that in high dose group ( P 〈 0.05 ). Conclusion The contents of AQP4,AQP8 increased obviously in colon tissue in the mice with constipation, Qilang-Heji could down-regulate the expression of AOP4 and AOP8.
