Research of correlation between plasma miRNAs and coronary heart disease

摘要 目的探讨血浆微小核糖核酸(miRNAs)在冠心病患者和健康人群中的表达差异及其对冠心病的诊断价值。方法收集102例冠心病患者(患者组,包括急性心肌梗死69例、心绞痛33例)和64例健康体检者(对照组)血浆标本,提取血浆总RNA,以U6sn RNA为内参采用实时荧光定量法分别检测miR-503、miR-186和miR-335的表达水平,并进行比较分析,通过受试者工作曲线(ROC)分析目标miRNAs对冠心病的诊断效能。结果急性心肌梗死组和心绞痛组患者血浆miR-503和miR-186表达水平均明显高于对照组(P均<0.05),而miR-335表达水平与对照组比较差异无统计学意义(P>0.05);急性心肌梗死组患者血浆miR-503、miR-186表达水平均明显高于心绞痛组(P均<0.05)。ROC分析显示miR-503的曲线下面积(AUC)为0.689(P<0.05),miR-186的AUC是0.679(P<0.05)。结论冠心病患者血浆中miR-503和miR-186表达水平显著升高,二者对冠心病有一定的诊断价值,有可能成为诊断冠心病的潜在生物标志物。 Objective It is to explore whether plasma miRNAs expression in coronary heart disease (CHD) patients differed from healthy controls and the potential value to serve as a biomarker for CHD diagnosis. Methods 102 cases of patients with CHD disease (patient group, including 69 cases of acute myocardial infarction and 33 cases of angina peetoris) and 64 healthy check-up individuals (eontrol group) were recruited and their plasma total RNA was extracted by using Qiagen kits. The expressions of miR-503, miR-186, miR-335 were detected by real time quantitative PCR method. Then a comparision for different miRNAs expression levels was carried out between the case and control groups. The diagnostic performance of target miRNAs for CHD was evaluated by Receiver Operating Characteristic (ROC). Results There was significant difference in the expression levels of miR-186 and miR-503 (P 〈 0.05) between AMI, AP groups and healthy control group, but the results of miR-335 showed no statistical difference ( P 〉 0.05 ). There was significant difference between AMI and AP groups ( P 〈 O. 05 ). The combined ROC analysis of miR-503 revealed an AUC value of 0. 689 ( P 〈 0.05 ) and an AUC value of 0. 679 ( P 〈 0. 05) for miR-186. Conclusion Increased plasma miR-186 and miR-503 in CHD patients strongly correlated with lipids. By the ROC analysis, miR-503 and miR-186 were of significant diagnostic value for CHD. In conclusion, these results indicated that they may serve as the potential biomarker for the diagnosis of CHD.
