Optimization of Processing Technology of Pinelliae rhizome by Box-Behnken Design Response Surface Methodology

摘要 目的研究法半夏炮制工艺的关键影响因素,优选法半夏炮制工艺。方法在单因素试验的基础上,以水溶性浸出物、鸟苷、甘草酸二铵含量的总评归一值(OD)为指标,对浸泡时间、干燥温度及干燥时间这3个因素进行响应面试验研究,从而得出最佳炮制工艺。结果优选最佳工艺条件为生半夏与甘草提取液和生石灰浸泡3天,于72℃干燥24h。该炮制工艺条件所测的实际值与理论值基本一致,偏差较小。结论通过响应面法优化法半夏炮制工艺,简便易行,结果可靠,能为法半夏质量控制和规模化生产提供科学依据和新颖思路。 Objective To study the key factors of processing technology of Rhizome Pinelliae and to optimize it by Box-Behnken design response surface methodology. Methods On the basis of single factor screening,Box-Behnken experimental design was employed,the overall desirability,which was based on the content of water soluble extract,guanosine,and diammonium glycyrrhizinate,were used as evaluation index. The soaking temperature,drying temperature,drying duration were used as independent variables. Results The optimal processing conditions were as follows: crude Rhizome Pinelliae was soaked in liquid extraction of Radix Glycyrrhizae,with calcium oxide,for 3 days,and dried at the temperature of 72℃ for 24 hours. The actual values determined by the response surface methodology are basically consistent with the theoretical values, with little deviation.Conclusion The optimized processing technology is easy to operate and the results are accurate and reliable,which could provide scientific basis and innovative idea for quality control and large-scale production of Rhizome Pinelliae.
