丙泊酚麻醉对创伤性脑损伤大鼠神经元再生和神经功能的影响 被引量:3

Effects of propofol on neuronal regeneration and neurological function after traumatic brain injury in rats

摘要 目的观察丙泊酚麻醉对创伤性脑损伤大鼠(TBI)的影响。方法用Feeney法建立TBI模型。按照体重将雄性Wistar大鼠分为4组:假手术组、模型组和低高2个剂量实验组,每组20只。低高2个剂量实验组尾静脉注射丙泊酚36,72 mg·kg^(-1)·h^(-1)。术后28 d,比较4组大鼠术后的死亡率,用平衡测验方法检测平衡能力,用巴恩斯迷宫法检测学习记忆能力,用免疫组化法检测神经元新生状况。结果术后28 d,低高2个剂量实验组大鼠死亡率分别为30%,40%,与模型组比较差异均有统计学意义(均P<0.05)。高剂量实验组大鼠进入目的箱时间为(169.64±6.77)s,显著高于模型组的(144.21±8.52)s,差异有统计学意义(P<0.05);高剂量实验组平衡能力为(37.29±4.26)s,显著低于模型组的(49.37±5.17)s,差异有统计学意义(P<0.05)。模型组大鼠的神经元新生数为(680.46±65.17)个,低高2个剂量实验组大鼠分别为(364.59±76.21),(302.14±42.77)个,与模型组比较差异均有统计学意义(均P<0.01)。结论丙泊酚可降低创伤性脑损伤大鼠神经元再生,且对创伤性脑损伤大鼠具有恶化作用。 Objective To observe the effect of propofol anesthesia on neuronal regeneration and neurological function after traumatic brain injury( TBI) in rats. Methods TBI model was established by Feeney method. Rats were divided into sham operation group,model group,low dose propofol group( 36 mg·kg^(-1)·h^(-1)) and high dose propofol group( 72mg·kg^(-1)·h^(-1)),n = 20 in each group. Propofol was administered intravenously from 30 min before to 1. 5 h after surgery. The mortality,neurological function and neuronal regeneration were determined. The postoperative mortality was compared between the 4 groups at 28 d after operation. The balance test was used to detect balance. The ability of learning and memory was tested by Barnes maze. The neuronal status was detected by immunohistochemistry. Results Twenty-eight days after operation,mortality in low dose propofol group and high dose group was respectively 30%,40%,significantly higher than model group and sham group( P〈0. 05). The time to correct box time in propofol high dose group was( 169. 64 ± 6. 77) s,which was significantly higher than that of( 144. 21 ± 8. 52) s in model group( P〈0. 05). The balance ability ofthe high dose group( 37. 29 ± 4. 26) s was significantly lower than that the model group( 49. 37 ± 5. 17) s( P〈0. 05).Whereas the propofol low dose group and high dose group could significantly decrease the neuronal regeneration( P〈0. 05). Conclusion Propofol can reduce neuronal regeneration after traumatic brain injury in rats,and it has a worsening effect on traumatic brain injury in rats.
