摘要
目的提取桑叶多糖并对分级醇沉所得桑叶多糖进行含量测定与单糖组成分析。方法用水提醇沉法提取桑叶多糖,以三氯乙酸除蛋白、活性炭脱色,用AB-8大孔吸附树脂纯化,以95%乙醇将桑叶多糖分级醇沉为四段(a、b、c、d);以苯酚-硫酸法测定桑叶多糖含量;用薄层色谱法与气相色谱法测定四段多糖中单糖组成。结果分级醇沉的多糖含量分别为38.4%,45.5%,40.7%,42.3%。在四段多糖中均含有3种已知单糖:鼠李糖、半乳糖、葡萄糖;部分含有木糖、甘露糖、果糖。结论在桑叶多糖中,不同分子量段的含量及其各单糖数目、种类和含量均有所区别。
Objective To extract of Polysaccharides from mulberry leaves( PML) and to determinate the content of PML derived from alcohol precipitation and analyze the composition of monosaccharide.Methods PML were extracted by water and precipitated by alcohol,proteinum eliminated by trichloroacetic acid,active carbon decolor,AB-8 macroporous resin purification. The PML graded alcohol precipitation into four groups a,b,c,d by 95% ethanol. The phenol-sulfuric acid method for determination the content of PML. The monosaccharide composition in four group Polysaccharides were determined using thin-layer chromatography and gas chromatography method. Results The content of Polysaccharides in four group by graded alcohol precipitation were 38. 4%,45. 5%,40. 7%,42. 3% respectively. The four group Polysaccharides contain three kinds of known monosaccharides,rhamnose,galactose and glucose; while the section contains xylose,mannose,fructose. Conclusion The content of different molecular weight segments and the number and types of monosaccharide in leaves of mulberry leaves are different.
