蒙药额力根-7对急性肝损伤小鼠模型影响的研究 被引量:7

摘要 目的:考察蒙药额力根-7对急性肝损伤的保护作用和急性毒性反应,为临床合理用药提供药理学参考.方法:(1)健康昆明种小鼠60只,随机分为6组,每组10只:分别为空白组,CCl_4模型组,水飞蓟素阳性对照组,额力根-7低剂量组、中剂量组、高剂量组.将适应环境的空白组和模型组小鼠给予生理盐水,药物组连续给予相应浓度的受试药物.10天后,除空白组给予0.1ml/10g麻油外,其他组均给予0.35%CCl_4麻油溶液0.1ml/10g制造小鼠肝损伤模型,24h后取血检测血清中AST、ALT、TNF-α的水平.处死小鼠,计算肝脏指数,HE染色,观察肝脏组织,并检测肝脏匀浆中MDA、SOD含量.(2)健康昆明种小鼠40只,随机分为2组,额力根-7组和空白组,每组20只:额力根-7组给予1.0 g/ml、体重为0.5ml/10g药物,空白组给予蒸馏水0.5ml/10g,连续观察14天,计算小鼠最大耐受量倍数.结果:(1)药物组与模型组比较,血清中AST、ALT、TNF-α水平明显下降(P<0.01);染色观察显示,额力根-7组肝脏的炎性浸润和肝细胞的肿胀程度较轻;药物组与模型组相比,肝脏指数和MDA下降明显(P<0.01),SOD明显升高(P<0.01).(2)小鼠最大耐受量为50g/kg.结论:额力根-7对CCl_4诱导的小鼠急性肝损伤具有保护作用. Objective: Through investigating the protective effect of Eligen-7 on acute mice liver injury and its acute toxicity,to provide a pharmacology reference for rational clinical application. Methods: 1.Sixty healthy Kunming mice were randomly divided into six groups: normal control group,CCl_4 model group,silymarin group and Eligen-7 low-,middle-,high-dose groups,10 mice in each group. The mice in the normal control group and the CCl_4 model group were given normal saline; meanwhile,the mice in drug group were given corresponding concentration drugs for 10 days. Except the normal control group,the other 5 groups were given 0.35% CCl_4,0.1 ml/10 g sesame oil to make the model of acute liver injury. Determinations of serum AST,ALT,TNF-α and pathological examination in all mice were carried out after 24 hours. At the same time,the SOD and MDA in liver homogenate were detected and the liver index was calculated. 2. Forty healthy Kunming mice were randomly divided into two groups: Eligen-7 groups and normal control group,20 mice in each group. The Eligen-7 groups were given 1.0 g/ml,0.5 ml/10 g Eligen-7,and the normal control group was given 0.5 ml/10 g distilled water for 14 days to test the maximum tolerance dose. Results: 1. Compared with the model control group,the serum levels of AST,ALT and TNF-α were remarkably decreased(P 0.01)in drug groups. Eligen-7 can significantly decrease inflammatory cell infiltration and swelling.The MDA decreased(P 0.01)and SOD clearly increased(P 0.01)in the drug groups. 2. The maximum tolerance dose to mice was 50 g/kg. Conclusion: Eligen-7 has protective effects on acute liver injury induced by CCl_4.
