摘要
目的:观察丹参注射液联合重组人粒细胞集落刺激因子(rhG-CSF)对急性脑梗死大鼠双侧海马齿状回(DG)干细胞(stem cells)增殖的影响。方法:采用大脑中动脉线栓法制作左侧大脑中动脉闭塞(MCAO)脑梗死大鼠模型,并随机分为丹参注射液组、rhG-CSF组、rhG-CSF+丹参注射液组、生理盐水组,每组6只。按实验方案给药后,每组在干预前及干预7 d后,采用改良神经功能损害评分(m NSS)综合评价神经系统损伤的严重程度;另随机选择6只正常大鼠作为假手术组。以免疫荧光染色法检测DG区与增殖细胞相关的Ki67(Ki67)/巢蛋白(nestin)、造血干细胞(CD34+)阳性细胞数。结果:rhG-CSF+丹参注射液组大鼠m NSS评分低于丹参注射液组、rhG-CSF组、生理盐水组,差异均有统计学意义(P<0.05);rhG-CSF+丹参注射液组DG的Ki67+/nestin+细胞数高于其余4组。rhG-CSF组及rhG-CSF+丹参注射液组在脑梗死侧均发现CD34+。结论:丹参注射液联合rhG-CSF能促进脑梗死大鼠海马齿状回神经新生,改善脑梗死大鼠的神经功能,可能在脑梗死后神经功能恢复过程中发挥了重要作用。
Objective: To observe the effect of Danshen injection and rhG-CSF on on the bilateral Dentate gyms (DG) promote the proliferation of stem cells in rats post cerebral infarction. Methods: The string fastenings method for middle cerebral artery occlusion (MCAO) rat models were randomly devided into Danshen injection group, rhG-CSF mobilization group, rhG-CSF mobilization+Danshen injection group and saline group, 6 rats in each group. Comprehensive evaluation the neurological deficits using the modified neurological severity score (mNSS) on each group before treatment and on the Th day after used the drugs. Another 6 normal rats were randomly selected as sham operation group. Ki67+/nestin+, CD34 +cells in the DG were detected by immunofluo- resccnce techniques. Results: RhG-CSF mobilization + Danshen injection group showed lower mNSS score than rhG-CSF mobilization group, Danshen injection group and saline group, and the difference was statistically significant (P〈O.05); The rhG-CSF mobilization + Danshen injection group showed more of Ki67+/nestin+ cells than the other four groups. There were CD34+ detected on the cerebral infarction side in rhG-CSF mobilization group and rhG-CSF mobilization +Danshen injection group. Conclusion: The combination of Danshen injection and rhG-CSF can promote the neurogenesis in Dentate gyrus of rats after cerebral infarction, which may a associate with the recovery the neural function.
