摘要
目的观察荞麦花叶发酵提取物(Extract of fermented buckwheat flower and leaf,EFBFL)对MIN6细胞促增殖,抗凋亡作用,为荞麦花叶进一步开发研究提供实验依据。方法以不同浓度EFBFL干预胰岛MIN6细胞,以高糖(含葡萄糖33.3 mmol/L)刺激作为诱导凋亡或抑制增殖因素。用cck-8试剂盒检测EFBFL对MIN6细胞增殖活性的影响;用流式细胞仪(Annexin V-FITC和PI双染法)检测EFBFL对胰岛细胞凋亡的影响。结果 EFBFL在16、32、64μg/ml浓度下对高糖和非高糖刺激的胰岛MIN6细胞增殖活性均有促进。与高糖对照组比较,32和64μg/ml的EFBFL对高糖诱导的胰岛细胞早期凋亡和晚期凋亡有明显抑制作用(P<0.01),有浓度效应关系。结论 EFBFL具有良好的促进MIN6细胞增殖和抑制高糖诱导的胰岛MIN6细胞的凋亡作用。
Objective To observe the effect of the extract of fermented buckwheat flower and leaf(EFBFL)on increasing cell proliferation activity and anti-apoptotic effect in MIN6 cells,and to provide experimental basis for the further development of EFBFL.Methods MIN6 cells were treated with various concentration of EFBFL to determine a suitable concentration of the cells to promote cell proliferation.Then,MIN6 cells were induced apoptosis or inhibited proliferation with glucose(33.3mmol/L).Cells proliferation activity was measured by CCK-8 kit.Effect of EFBFL on apoptosis in MIN6 cells was observed by Annexin V-FITC/PI staining immunofluorescence and was analyzed by flow cytometry.Results The proliferation activity of MIN6 cells was increased at the concentration of 16,32,64μg/ml in high glucose and non-high glucose stimulation.Compared with the high glucose group,EFBFL at 32 and 64μg/ml dosage had a significant inhibitory effect on high glucose-induced early apoptosis and late apoptosis of MIN6 cells(P〈0.01),and this effect had a concentration effect relationship.Conclusion EFBFL has a good effect on increasing the proliferation activity of MIN6 cells and inhibiting the apoptosis of MIN6 cells induced by high glucose.
