摘要
目的观察椰子壳挥发油对急性心肌梗死大鼠心肌过氧化损伤的保护作用。方法选取肢体Ⅱ导联心电图正常的40只Wistar大鼠,随机分为四组,即假手术组、模型组、挥发油组、复方丹参滴丸组,各组给药均为1次/d,连续7d;采用冠状动脉左前降支结扎造成AMI大鼠动物模型,检测各组大鼠心功能,血清和心肌组织iNOS、eNOS、T-SOD活力及MDA含量。结果模型组射血分数(EF)、左室缩短率(FS)下降明显,与假手术组比较有差异(P<0.01);各用药组心功能指标改善明显,与模型组比较差异有统计学意义(P<0.01)。模型组大鼠iNOS升高,eNOS降低,与假手术组比有差异;各给药组与模型组比较,NO合成酶改善明显,挥发油组血清和心肌组织中iNOS和eNOS活力与模型组比较均差异有统计学意义;丹参滴丸组与挥发油组比较均差异无统计学意义(P>0.05)。模型组大鼠MDA含量升高,T-SOD活力降低,与假手术组比差异有统计学意义(P<0.01);与模型组比较,丹参滴丸组T-SOD和MDA均差异有统计学意义(P<0.01),椰子壳挥发油组T-SOD活力有差异(P<0.01),而MDA含量无统计学差异;丹参滴丸组与挥发油组比差异无统计学意义(P>0.05)。结论椰子壳挥发油可以减轻急性心肌梗死大鼠心肌的过氧化损伤,作用机制与稳定细胞膜,保护内源性抗氧化酶活性方面有关。
Objective To observe the protective effect of volatile oil of coconut shell on myocardial peroxida- tion of acute myocardial infarction rats.Methods Forty Wistar rats with limb Ⅱ lead electrocardiogram (ecg} normal were randomly divided into 4 groups, namely the control group, model group, naphtha, compound dan- shen dropping pill group,each group 1 times/d,continuous 7 d.The animal model of the AMI rats was caused by the ligation of left anterior descending coronary artery, and the cardiac function of rats in each group was detected, and iNOS, eNOS,T- SOD energy and MDA content in the serum and myocardial tissue were detec- ted.Results The decrease of the ejection fraction (EF) and the decrease of the left ventricle (FS) in the mod- el group were obvious, compared with the sham operation group (P 〈0.01).The improvement of cardiac func- tion in each medication group was significantly improved, compared with the model group {P〈0.01 ). The model group rats were elevated, the eNOS decreased, compared to the pseudosurgical group.Compared with the model group,NO synthase was improved,and the energy of iNOS and eNOS in the serum and myocardial tissues of the volatile oil group were different from that of the model group.There was no difference between danshen drop pill and volatile oil group (P〈0.05).The MDA content of rats in the model group increased and the activity of t- sod decreased compared with the sham operation group (P〈0.01).Compared with the mod- el group , the t-sod and MDA were different (P〈0.01),and the energy of T- SOD was different (P〈0. 01 ), and the MDA content was not statistically significant.There was no difference between danshen drop pill group and volatile oil group (P〈0.05).Conclusion The volatile oil of coconut shell can alleviate the peroxi- dative damage of myocardial infarction in acute myocardial infarction, and its mechanism is related to stable cell membrane and the protection of endogenous antioxidant activity.
