摘要
以柠檬酸为脱钙剂,研究搅拌条件下鲢鱼鱼鳞的脱钙条件,并通过扫描电子显微镜对脱钙前后鱼鳞的表面结构进行观察对比。采用单因素及正交试验,考察料液比、脱钙时间及柠檬酸浓度对鱼鳞脱钙效果的影响。结果表明,最佳工艺参数为:料液比1∶20(g/mL),脱钙时间120 min,柠檬酸浓度3.5%,在此条件下,脱钙率可达到95.23%,电镜结果显示,脱钙能使鲢鱼鱼鳞的蛋白质裸露出来,有利于鱼鳞蛋白质的提取。
The process conditions of citric acid removal the calcium ions from the silver carp fish-scale were studied. The surface structures of undecalcified fish scale and decalcified fish scale were observed by scanning electron microscope(SEM). The effects of solid-liquid ratio,reaction time and citric acid concentration on the decalcification of fish scales were investigated and the optimal technological parameters were separately to be 1 ∶20(g/mL),120 min and 3.5 %. Under the optimal conditions,decalcification rate was 95.23 %. SEM results showed that decalcification could be favorable for the extraction of fish scale protein.
