基于MT3、SOD2 mRNA表达调控的醒脑再造胶囊抗脑缺血再灌注损伤作用

摘要 目的观察醒脑再造胶囊的抗脑缺血再灌注损伤作用并探讨其作用机制。方法灌胃给药醒脑再造胶囊1周后,建立SD大鼠大脑中动脉缺血2 h再灌注24 h模型,进行大鼠神经功能评分,2,3,5-三苯基氯化四氮唑(TTC)染色法观察脑组织梗死范围,实时荧光定量PCR检测脑组织缺血半暗带区域金属硫蛋白3(MT_3)和超氧化物歧化酶2(SOD_2)mRNA的表达。结果醒脑再造胶囊预处理可明显降低大鼠脑缺血再灌注损伤后神经功能评分,缩小脑组织梗死范围,并使脑组织缺血半暗带区域MT_3和SOD_2 mRNA的表达显著增加。结论醒脑再造胶囊抗大鼠脑缺血再灌注损伤作用与上调脑组织抗氧化损伤基因MT_3和SOD_2 mRNA的表达有关。 Objective To observe the protective effects of Xingnaozaizao capsule( XNZZ) on cerebral ischemia reperfusion injury and explore the mechanism. Methods After intragastric administration of XNZZ for 7 days,the SD rat model of cerebral injury was established by 2 h ischemia and 24 h reperfusion. Effects of XNZZ on neurological scale were observed,the infarction volume of brain was detected by TTC staining,and the mRNA expressions of MT3,SOD2 were determined by real-time PCR. Results XNZZ can obviously reduce the neurological scale,decrease the infarction volume of brain,and increase the mRNA expressions of MT3,SOD2 of rats after cerebral ischemia reperfusion injury. Conclusion The protective effects of XNZZ on cerebral ischemia reperfusion injury would be associated with inducing the activation of the antioxidant genes MT3 and SOD2.
