摘要
目的分析马鞭草总黄酮对人肝癌细胞株Hep G-2增殖及IL-6、JAK2、STAT3 mRNA水平的影响。方法采用不同质量浓度的马鞭草总黄酮处理体外培养的肝癌Hep G-2细胞,Real time PCR检测IL-6、p-JAK2、p-STAT3表达水平,Western blot法分析JAK2、STAT3蛋白表达水平。结果与空白对照组比较,马鞭草总黄酮能显著降低IL-6、p-JAK2、p-STAT3 mRNA表达水平(均P<0.05),下调JAK2、STAT3蛋白水平(P<0.05)。结论马鞭草总黄酮下调肝癌Hep G-2细胞IL-6、JAK2、STAT3水平,抑制细胞增殖。
Objective To investigate the total flavonoids of Verbena officinalis L (TFV) effect on IL-6, JAK2,STAT3 level of human hepatocellular carcinoma HepG-2 ceils. Methods HepG-2 Ceils were treated and cultured with different concentrations of TFV. The expression of IL-6,p-JAK2, p-STAT3 mRNA were detected by RT-PCR. Expression of p-JAK2, p-STAT3 protein were analyzed by western blotting. Results Compared with the control group, TFV could significantly decrease the expressions of IL-6, p-JAK2, p-STAT3 (all P 〈 0.05) , Moreover, TFV could down-regulate the protein expression of p-JAK2, p-STAT3 (P 〈 0.05 ). Conclusion TFV could down-regulate the level of IL-6, JAK2, STAT3 and inhibit the proliferation of HepG-2 cells.
