莪术50%乙醇大孔树脂洗脱部位抗血栓作用及其机制研究 被引量:11

Study on the anti-thrombotic effect of the 50% ethanol eluting fraction from Curcuma kwangsiensis

摘要 目的:研究广西莪术大孔树脂50%乙醇洗脱部位的抗血栓作用,并探讨其作用机制。方法:采用角叉菜胶致小鼠尾部血栓模型,电刺激致大鼠实验性颈总动脉血栓模型,大鼠动-静脉旁路血栓模型,观察广西莪术50%乙醇大孔树脂洗脱部位抗血栓作用;用FeCl_3诱导大鼠体内血栓模型,观察其抗血栓作用及对大鼠NO、ET-1、6-keto-PGF1α、TXB_2等含量的影响;用大鼠血小板聚集实验观察其对血小板聚集的影响。结果:广西莪术50%乙醇洗脱部位(6.0、3.0、1.5 g生药/kg)能明显减少角叉菜胶所致小鼠尾部血栓黑尾动物数,减小黑尾长度;广西莪术50%乙醇大孔树脂洗脱部位(3.0、1.5、0.75 g生药/kg)明显延长电刺激致大鼠实验性颈总动脉血栓形成时间,明显减轻大鼠动-静脉旁路血栓湿重;明显减轻FeCl_3诱导体内血栓模型大鼠血栓湿重,增加大鼠NO、6-keto-PGF1α含量,降低ET-1、TXB_2含量,提高NO/ET-1、6-keto-PGF1α/TXB_2比值;能明显抑制血小板聚集。结论:广西莪术50%乙醇大孔树脂洗脱部位具有明显的抗血栓作用,其机制与调节NO、ET-1、6-keto-PGF1α、TXB_2水平和NO/ET-1、6-keto-PGF1α/TXB_2比值,抑制血小板聚集有关。 Objective: To investigate the anti-thrombotic effects of the 50% ethanol eluting fraction by macroporous resin from Curcuma Kwangsiensis and its mechanism. Methods: The mouse tail thrombosis induced by the carrageenan the,the rat common carotid artery thrombosis induced by the electric stimulation,the rat arteriovenous bypass thrombosis were used to observe the anti-thrombotic effects the 50% ethanol eluting fraction from Curcuma Kwangsiensis. The rat in vivo thrombus induced by FeCl_3 was used to observe the anti-thrombotic effect. Contents of NO,ET-1,6-keto-PGF1α,TXB_2 in rats were measured. The platelet aggregation text in rats were performed. Results: The 50% ethanol eluting fraction( 6. 0,3. 0,1. 5g crude drug/kg) significantly decreased the number of mice with the blackened tail induced by the carrageenan and the length of the blackened tail. The 50% ethanol eluting fraction( 3. 0,1. 5,0. 75 g crude drug/kg) significantly extended the thrombus formation time of rats with the common carotid artery thrombosis induced by the electric stimulation,significantly reduced wet weights of thrombus in arteriovenous bypass thrombosis and rats in vivo thrombus models,increased contents of NO,6-keto-PGF1α,decreased contents of ET-1,TXB_2,significantly increased ratios of NO/ET-1,6-keto-PGF1α/TXB_2; significantly inhibited the platelet aggregation in rats. Conclsion: The 50% ethanol eluting fraction from Curcuma Kwangsiensis has significantly anti-thrombotic effect. The mechanism may be involved in regulating levels of NO、ET-1,6-keto-PGF1α,TXB_2 and ratios of NO/ET-1,6-keto-PGF1α/TXB_2,the inhibitation of platelet aggregation.
