芪明颗粒对2型糖尿病肾病大鼠模型肾脏保护作用研究

Protective Effects of Qiming Granule on Experimental Type 2 Diabetic Nephropathy Rats

目的:观察芪明颗粒对2型糖尿病肾病大鼠模型肾脏保护作用研究,并探讨其治疗DN的作用机制。方法:选用SPF级SD大鼠60只,除空白组外各组均给予链脲佐菌素单次腹腔注射造模,随机分为空白组、模型组、低剂量组、高剂量组、缬沙坦组。8周后分别用Western blot法、ELISA法检测肾组织中WT1、ET-1、AngⅡ的表达。结果:模型组肾组织WT1、AngⅡ、ET-1明显高于空白对照组(P<0.05);与模型组相比,低剂量组、高剂量组、缬沙坦组肾脏组织WT1、AngⅡ、ET-1均显著降低(P<0.05);高剂量组、缬沙坦组肾组织WT1、AngⅡ、ET-1明显低于中药低剂量组(P<0.05),中药高剂量组、缬沙坦组相比差异无统计学意义(P>0.05)。结论:芪明颗粒可以有效降低DN大鼠肾组织WT1、AngⅡ、ET-1的表达,起到保护肾脏的作用,且高剂量芪明颗粒较低剂量对肾脏有更好的保护作用,可见早期采用芪明颗粒对糖尿病进行干预,可延缓肾损害的进展。

Objective：To observe the protective effect of Qiming Granule on renal function in rats with type 2 diabetic nephropathy（DN） and to study its action mechanism on DN.Methods：60 SD rats of SPF grade were selected and randomly divided into the blank control group,the model group,the low-dose group,the high-dose group and the Valsartan group.Streptozotocin（STZ） was used to set up the DN model in rats by intraperitoneal injection.After eight weeks,the expressions of WT1,ET-1 and AngII in renal tissues were detected by Western blot and ELISA methods respectively.Results：The level of WT1,AngII,ET-1 were significantly higher in the model group than those in the blank control group （P〈0.05）;and they were all significantly decreased in the low-dose group,in the high-dose group,and in the Valsartan group,when compared to the model group（P〈0.05）;the level of WT1,AngII,ET-1 both in the high-dose group and the Valsartan group were lower than those in the low-dose group（P〈0.05）,but there was no statistical significance between the high-dose group and the Valsartan group （P〉0.05）.Conclusion：Qiming granule can effectively reduce the expressions of WT1,AngII and ET-1 in renal tissue of DN rats,which can protect the kidney.High-dose Qiming granule has better protective effect on kidney.Early intervention can delay the progression of renal damage.
