摘要
目的:建立不同产地古丽娜的HPLC指纹图谱,考察不同产地古丽娜药材的质量。方法:采用Venusil XBP C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-0.4%磷酸溶液(B),梯度洗脱,流速为1.0 mL/min;柱温:25℃;检测波长:280 nm;进样量10μL。结果:确定由12个共有峰构成古丽娜药材的指纹特征,精密度和重复性中共有峰相对保留时间和相对峰面积RSD均小于3.0%,12批样品与生成的对照指纹图谱的相似度均大于0.85。对12批药材进行聚类分析,分成3类,聚类分析显示产地和采集时间与药材的质量相关。结论:该方法精密度高、重现性好,可为古丽娜药材的质量评价提供参考,也为古丽娜药材进一步开发利用奠定基础。
Objective : To establish the chromatographic fingerprint of different origin of Gulina medicine, in order to investigate the quality of Gulina medicinal materials in different origin. Methods:The determination was carried out on a Venusil XBP C18 (250 mm x 4. 6 mm ,5 μm)column, a gradient elution of acetonitrile( A) -0. 4% phosphoric acid(B) solution as the mobile phase, the flow rate was 1.0 mL/min, the column temperature was at 25 %, the detection wavelength was 280 nm, the injection volume was 10 μL. Results:The fingerprint features of Gulina medicine were determined by twelve common peaks. The relative retention time and relative peak area of RSD were all less than 3.0%, and the similarity of the twelve batches of samples in comparision with the control figerprint were more than 0. 85. Cluster analysis of the twelve batches of Gulina medicinal herbs, which divided into three categories, the clustering analysis result showed that the origin and collection time influenced the quality of medicinal herbs. Conclusion:The method has high precision and good reproducibility,which can provide reference for the quality evaluation of Gulina medicine, and lay the foundation for further development and utilization of Gulina medicine.
