亲水作用色谱法同时测定保健品中的硫酸软骨素和氨基葡萄糖 被引量:1

Simultaneous determination of chondroitin sulfate and glucosamine in functional foods by hydrophilic interaction liquid chromatography

摘要 目的建立亲水作用色谱结合PDA检测器同时测定保健品中硫酸软骨素和氨基葡萄糖的高效液相色谱法。方法采用Waters Xbridge HILIC色谱柱(4.6 mm×250 mm,5μm),以乙腈-10 mmol/L乙酸铵(95∶5,V/V)为流动相,流速为1.0 ml/min,柱温为35℃,检测波长为192 nm,进样体积为5μl。结果在上述色谱条件下,硫酸软骨素和氨基葡萄糖在10μg/ml^200μg/ml内线性关系良好,硫酸软骨素和氨基葡萄糖的检出限均为3.0μg/ml,回收率分别为94.18%、93.45%。混合溶液在48 h内稳定。结论本方法经方法学检验,灵敏度和专属性高,稳定可靠,适用于同时测定保健品中的硫酸软骨素和氨基葡萄糖。 Objective To establish a method for simultaneous determination of chondroitin sulfate and glucosamine in functional foods by hydrophilic interaction liquid chromatography( HILIC) coupled with photo-diode array detector. Methods The sample was analyzed on a Waters Xbridge HILIC column( 4. 6 mm × 250 mm,5 μm),with the mobile phase of acetonitrile-10 mmol/L ammonium acetate solution( 95∶5,V/V) at the flow rate of 1. 0 ml/min and the column temperature was 35 ℃;the detection wavelength was 192 nm and the injection volume was 5 μl. Results The method showed a good linearity when the concentrations of chondroitin sulfate and glucosamine were within 10 μg/ml-200 μg/ml. Both detection limits of chondroitin sulfate and glucosamine were 3. 0 μg/ml and the recoveries were 94. 18% and 93. 45%,respectively. The solution was stable in 48 h. Conclusion This verified method had the advantages of high sensitivity,high specificity and high reliability,which is suitable for the simultaneous determination of chondroitin sulfate and glucosamine in functional foods.
