摘要 目的:建立气道炎症模型豚鼠血浆中花生四烯酸(AA)的测定方法,考察野鸢尾黄素降低气道炎性反应的作用。方法:以烟熏配合合胞病毒滴鼻制造气道炎症豚鼠模型,以AA-d8为内标,采用Triton-14对血浆样品进行浊点萃取,并采用LC-MS/MS法MRM监测模式测定豚鼠血浆中AA的浓度。结果:血浆中AA在0.1~100μg·m L-1范围内线性关系良好,日内精密度为2.53%~7.82%,日间精密度为10.51%~13.41%,提取回收率81.20%~90.77%。结论:该方法环保、准确、稳定,可用于豚鼠血浆中AA的测定。 Objective: To establish a method for the determination of arachidonic acid( AA) in plasma of guinea pigs with airway inflammation model and investigate the effect of irigenin on reducing airway inflammatory response. Methods: The model of airway inflammation in guinea pigs was made by smokation combined with nasal infection. The concentration of AA in guinea pig plasma was determined by LC-MS/MS with MRM monitoring mode by using Triton-14 as the cloud point extraction and AA-d8 as internal standard. Results: The linearity of AA in the range of 0. 1-100 μg·m L-1was good. The inter-day precision was2. 53% ~ 7. 82%. The intra-day precision was 10. 51% ~ 13. 41%. The recovery was 81. 20% ~ 90. 77%. Conclusion: The method is environmentally friendly,accurate and stable and can be used for the determination of AA in guinea pig plasma.
