藏药诃子的质量标准提高研究 被引量:11

摘要 目的:提高诃子的质量标准。方法:以没食子酸、诃子对照药材为对照,新增薄层色谱-生物自显影鉴别;建立HPLC测定没食子酸含量的方法,采用Phenomenex luna C18色谱柱(250mm×4.6mm,5μm);流动相为甲醇-0.1%磷酸溶液(5:95);流速1.0 m L/min;柱温30℃;检测波长271 nm;进样量10μL。结果:薄层色谱-生物自显影法鉴别斑点清晰,分离度好,不仅能对诃子进行定性鉴别,还能体现其抗氧化活性;在以上HPLC条件下,没食子酸在10.72~85.76μg呈良好线性关系,线性回归方程为Y=31.248X+9.037(r=0.9997),平均回收率96.1%(RSD 2.54)。结论:新增的方法简单易行,专属性强,重复性好,可用于诃子的质量控制。 Objective To improve the quality standards of Terminalia chebula Retz. Methods New additions to TLC - Bioautogra- phy method is based on gallic acid and control herbs as contrast, and combined with traditional TLC identification method for rapid qualitative identification of Terminalia chebula Retz. The contents of gallic acid was determined by HPLC. Gallic acid were separated on a phenomenex luna C18 column (250mm ×4. 6mm, 5μm ), using methanol -0. 1% phosphoric acid (5: 95) as the mobile Phase at a flow rate of 1.0 mL/min. The column temperature was 30 ℃ and the detection wavelength was 271 nm. The injection volume is 10 ixL. Results The spots in TLC were clear and well - separated. Not only on the Terminalia chebula Retz for qualitative identification, but also reflect its antioxidant activity. The methodology validation for the assay of gallic acid presented that it was in good linear correlation in the ranges of 10. 72 - 85.76 txg with the regression equations of Y = 31. 248X + 9. 037 ( r = 0. 9997) , and the average recoveries were 96. 1% (RSD 2. 54% ) . Conclusion These methods are simple, easy, strongly specific and good repeatability, which can be used for the quality control of Terminalia chebula Retz.
