改良Lowry法测定不同产地凹纹胡蜂药材蛋白质含量

Study on An Improved Lowry Method for Detecting the Content of Protein in Vespa velutina auraria Smith

目的:建立凹纹胡蜂药材中蛋白质含量测定的方法。方法:采用改良Lowry法测定标准小牛血清蛋白(BSA)和凹纹胡蜂药材中的蛋白质含量并对其进行方法学考察。结果:该方法线性及相关度良好,线性范围较宽,精密度、稳定性、重复性和加样回收率较佳;用该方法分别对不同产地的3批凹纹胡蜂药材中蛋白质含量进行测定,蛋白质含量分别为:德宏州(批号:20150630)12.37%,保山市(批号:20150927)13.08%,曲靖市(批号:20151112)9.63%,药材含水量经测定依次为6.12%、9.92%、10.17%。结论:不同产地凹纹胡蜂药材中蛋白质含量有一定差异,采自曲靖市的样品蛋白质含量偏低;实验采用的改良Lowry法测定结果准确、简便、稳定,适用于凹纹胡蜂药材中蛋白含量的测定。

Objective To establish a method for the content determination of protein in Vespa velutina auraria Smith. Methods The concentrations of bovine serum albumin （ BSA ） and V. velutina auraria was determined by the modified Lowry method, and to carry out methodological study. Results This method had good correlation linearity, wide linear range and good precision, stability, repeatability and recoveries. Conclusion The modified Lowry method to determine the protein concentration of V. velutina auraria is accurate, simple and stability, it＇s suitable for protein content determination of V. velutina auraria.
