摘要
目的探讨瞬时受体电位香草酸受体通道1(TRPV1)在大鼠宫颈扩张痛中的作用。方法成年雌性大鼠60只,体重200-250g,除对照组外,其余大鼠分别给予25kg、50kg砝码进行持续10s宫颈扩张(CD),重复1次,间隔5min,对照组不进行宫颈扩张。采用随机数字表法分为5组(n=12):对照组(C组)、CD25组(H1)、CD50组(H2)、CD25+鞘内注射Capsazepine组(CH1组)、CD50+鞘内注射Capsazepine组(CH2组)。分别记录各组宫颈扩张前(T_0)及宫颈扩张刺激后腹直肌肌电活动变化值。待UCD结束后10min,取胸腰段脊髓组织,采用Western Blotting法测定TRPV1表达。结果与C组比较,宫颈扩张刺激后H1组和H2组肌电活动明显增加,脊髓TRPV1表达上调(P<0.05);与H1组比较,宫颈扩张刺激后H2组肌电活动和脊髓TRPV1表达明显增加(P<0.05)。与H1、H2组比较,鞘内注射Capsazepine后两组肌电活动和脊髓TRPV1表达明显降低(P<0.05)。结论瞬时受体电位香草酸受体通道1可能参与了大鼠子宫颈扩张痛的形成。
Objective Investigate the role of transient receptor potential receptor 1(TRPV1) in cervical dilation pain in rats.Method 60 adult female rats,weighing 200-250 g,except the control group,the rats were given 25 kg and 50 kg weights for 10 s cervical dilatation(CD),repeated 1 times,interval 5min,control group without cervical dilatation. Methods the patients were randomly divided into 5 groups(n=12):control group(group C),group CD25(H1),group CD50(H2),CD25+ intrathecal injection,group Capsazepine(group CH1),and CD50+ intrathecal injection group Capsazepine(group CH2). The changes of EMG activity of rectus abdominis muscle before and after cervical dilation(T_0) and dilatation of cervix were recorded respectively. After UCD was finished,10 min was taken from the spinal cord of thoracolumbar spine and the expression of TRPV1 was measured by Western Blotting. Result Compared with the C group,the EMG activity of the H1 and H2 groups increased significantly and the expression of TRPV1 in the spinal cord increased after the cervical distension stimulation(P〈 0.05);Compared with group H1,the activity of EMG and the expression of TRPV1 in spinal cord of H2 group increased significantly after cervical distension stimulation(P〈0.05);Compared with group H1 and group H2,the activity of EMG and the expression of TRPV1 in spinal cord of two groups were significantly lower after intrathecal injection of Capsazepine(P〈0.05). Conclusion Transient receptor potential,1,may be involved in the formation of cervical dilatation pain in rats.
