HBD1对新生儿脐带血CD4^+T淋巴细胞活化和增殖的研究

Effect of HBD1 on activation and proliferation of CD4^+T lymphocytes in umbilical cord blood of neonates

本文探讨了HBD1对脐血CD4^+T细胞的增殖、细胞表面分子CD69、CD25、CD40L和CD45RO的表达及B细胞CD80、CD86分子表达的影响,并以成人外周血CD4^+T细胞作对照。采用CCK-8增殖实验检测CD4^+T细胞的增殖情况,采用流式细胞术检测CD80、CD86、CD69、CD25、CD45RO和CD40L的表达情况。结果表明:HBD1促进了脐血CD4^+T细胞的增殖,CD69、CD25、CD40L表达上调;B细胞CD80、CD86的表达也上调,但并未促进CD45RO表达的上调。而相同处理条件下,HBD1对成人外周血CD4^+T细胞作用不明显,提示HBD1对脐血来源CD4^+T细胞具有一定的免疫调节作用。

This study was to explore the effect of HBD1 on the proliferation of umbilical cord blood CD4~＋T cells and their expressions of CD69,CD25,CD40 L,CD45RO,CD80 and CD86.Adult peripheral blood was used as the control.The proliferation of CD4~＋T lymphocytes was detected by CCK-8,and the expressions of CD80,CD86,CD69,CD25,CD40 L,CD45RO were detected by FACS.The results showed that HBD1 promoted the proliferation of CD4~＋T cells and their expressions of CD69,CD25,CD40 Las well as the expressions of CD80/CD86 on B cells.However,HBD1 did not promote the expression of CD45 RO,which suggests HBD1 has an immunoregulatory effect on cord blood CD4~＋T cells.
