不同沙棘果渣添加水平对公绵羊睾丸和附睾抗氧化性的影响 被引量:5

摘要 [目的]研究饲喂不同沙棘果渣添加水平的日粮对公绵羊睾丸与附睾抗氧化性的影响。[方法]选用24只5月龄,体重(25.00±1.00)kg健康的杜泊×小尾寒羊杂交公羔,随机分为4组,按照分组依次饲喂添加不同水平沙棘果渣(0%、10%、20%、30%)的日粮,试验期65d。试验结束后采集公绵羊的睾丸和附睾组织,并对组织抗氧化相关基因mRNA和蛋白的表达进行研究。[结果]睾丸组织中,30%沙棘果渣添加组GPx4 mRNA的相对表达量相较于10%添加组以及对照组显著降低(P<0.05)。附睾头组织中,30%沙棘果渣添加组Cu-ZnSOD mRNA的相对表达量显著低于10%和20%沙棘果渣添加组(P<0.05);30%沙棘果渣添加组GPx4 mRNA的相对表达量相较于对照组显著降低(P<0.05)。附睾体组织中,30%沙棘果渣添加组CAT mRNA的相对表达量相较于10%添加组显著降低(P<0.05)。附睾尾组织中,20%沙棘果渣添加组Cu-ZnSOD mRNA的相对表达量相较于30%添加组与对照组显著升高(P<0.05);30%沙棘果渣添加组Cu-ZnSOD mRNA的相对表达量相较于10%添加组显著降低(P<0.05)。20%沙棘果渣添加组附睾头Nrf2与附睾尾Cu-ZnSOD蛋白表达量显著高于10%和30%沙棘果渣添加组。[结论]日粮中添加10%和20%沙棘果渣可以不同程度促进公绵羊睾丸和附睾抗氧化相关基因mRNA的表达,而添加30%沙棘果渣会降低睾丸GPx4、附睾头Cu-ZnSOD和GPx4、附睾体CAT、附睾尾Cu-ZnSOD mRNA以及附睾头Nrf2和附睾尾Cu-ZnSOD蛋白的表达量。因此,25kg杜泊×小尾寒羊杂交公羔日粮中添加30%沙棘果渣为过量添加水平。 [Objective]The purpose of this study was to investigate the effects of dietary Sea buckthorn Pomace on antioxidation in testis and epididymis of ram.[Methods]A total of 24Dopor×Small Tail Han Sheep ram lambs(5month old,25.00±1.00kg)were randomly selected and equally divided into 4groups,and received diet containing different level of sea buckthorn pomace(0%,10%,20%,30%).At the end of the trial(65d),the samples of testis and epididymis were collected and the mRNA and protein expression were detected.[Results]The results were as follows:The mRNA expression of GPx4 in testis was significantly lower in 30% group than that in control group and 10% supplementation group(P 0.05).Compared with the 10% and 20% groups,the mRNA expression of Cu-ZnSODin 30%group was significantly lower in caput epididymidis(P0.05).The mRNA expression of GPx4 in caput epididymidis was significantly lower in 30%supplementation group than this in control group(P0.05).Compared with the group with 10% sea buckthorn pomace,the mRNA expression of CATin 30% group was significantly lower in corpus epididymidis(P0.05).The mRNA expression of Cu-ZnSODin cauda epididymidis was significantly lower in control group and 30% group than that in 20% sea buckthorn pomace supplementation group(P0.05).In addition,com-pared with 10%sea buckthorn pomace addition group,the mRNA expression of Cu-ZnSODin 30% group was significantly lower in cauda epididymidis(P0.05).The protein expression of Nrf2 in caput epididymal and Cu-ZnSOD in the cauda epididymal were significantly higher in 20% group than that in 10% group and 30% group(P0.05).[Conclusion]The results indicated that supplementation of 10%and 20% of dietary sea buckthorn pomace could improve the mRNA expression of antioxidation related genes expression in testis and epididymis of ram,whereas supplementation of 30% sea buckthorn pomace reduced the mRNA expression of GPx4 in testis,Cu-ZnSODand GPx4 in caput epididymidis,CATin corpus epididymidis and Cu-ZnSODin cauda epididymidisa and the protein expression of Nrf2 in caput epididymidis and Cu-ZnSOD in cauda epididymidisa.Therefore,dietary 30% sea buckthorn pomace is not optimal.
