凋亡诱导剂TWEAK及细胞外基质蛋白Del-1与胰腺癌恶性程度的相关性研究

摘要 目的:研究凋亡诱导剂TWEAK及细胞外基质蛋白Del-1与胰腺癌恶性程度的相关性。方法:选择接受根治性切除手术的胰腺癌患者54例以及接受部分性切除手术的胰腺外伤患者32例,分别收集胰腺癌组织和正常胰腺组织适量,抽提RNA并反转录为cDNA后采用荧光定量PCR试剂盒测定TWEAK、Del-1、Survivin、USP9X、DPF2、Stathmin、CD44v9、GPSM2、MMP9、Snail、Vimentin的mRNA表达量。结果:胰腺癌组织中TWEAK、Del-1、Survivin、USP9X、DPF2、Stathmin、CD44v9、GPSM2、MMP9、Snail、Vimentin的mRNA表达量显著高于正常胰腺组织;TWEAK高表达胰腺癌组织中Survivin、USP9X、DPF2、Stathmin的mRNA表达量显著高于TWEAK低表达胰腺癌组织;Del-1高表达胰腺癌组织中CD44v9、GPSM2、MMP9、Snail、Vimentin的mRNA表达量显著高于Del-1低表达胰腺癌组织。结论:胰腺癌组织中TWEAK和Del-1的高表达分别能够促进胰腺癌细胞的增殖和侵袭。 Objective:To study the correlation of apoptosis inducer TWEAK and extracellular matrix protein Del-1with pancreatic cancer malignancy.Methods:A total of 54 patients with pancreatic cancer who underwent radical resection and 32 patients with pancreatic trauma who underwent partial resection were selected,proper amount of pancreatic cancer tissue and normal pancreatic tissue were collected respectively,RNA was extracted and synthesized into cDNA by reverse transcription,and then fluorescence quantitative PCR kits were used to determine TWEAK,Del-1,Survivin,USP9 X,DPF2,Stathmin,CD44v9,GPSM2,MMP9,Snail and Vimentin mRNA expression.Results:TWEAK,Del-1,Survivin,USP9 X,DPF2,Stathmin,CD44v9,GPSM2,MMP9,Snail and Vimentin mRNA expression in pancreatic cancer tissue were significantly higher than those in normal pancreatic tissue;Survivin,USP9 X,DPF2and Stathmin mRNA expression in pancreatic cancer tissue with high TWEAK expression were significantly higher than those in pancreatic cancer tissue with low TWEAK expression;CD44v9,GPSM2,MMP9,Snail and Vimentin mRNA expression in pancreatic cancer tissue with high Del-1expression were significantly higher than those in pancreatic cancer tissue with low Del-1expression.Conclusion:The high expression of TWEAK and Del-1in pancreatic cancer tissue can promote the proliferation and invasion of pancreatic cancer cells respectively.
