苍术白虎汤干预急性痛风性关节炎大鼠的疗效及机制探讨

Exploration of Efficacy and Mechanism of Rhizoma Atractylodis-White Tiger Decoction(苍术白虎汤) on Acute Gouty Arthritis in Rats

目的:观察苍术白虎汤对大鼠急性痛风性关节炎(acute gouty arthritis,GA)的疗效并探讨其机制。方法:36只大鼠分为苍术白虎汤高、中、低剂量组,西药组,模型组和对照组。建立大鼠急性痛风性关节炎模型,对照组注射等量生理盐水。以缚线法测量右侧踝关节同一部位周径(取均值),评估其关节肿胀度;于造模72 h后腹腔静脉采血,ELISA法测定其血清IL-1b、TNF-α水平,对血浆白细胞进行计数。结果:苍术白虎汤可显著降低急性痛风性关节炎大鼠关节肿胀度,降低其血清IL-1b、TNF-α水平及血浆白细胞,以高剂量组最为明显(P<0.01)。结论:苍术白虎汤可通过降低IL-1b与TNF-α表达、抑制白细胞增生而发挥较好的抗急性痛风性关节炎作用。

Objective： To observe the curative effect of Rhizoma atractylodis-white tiger decoction against rats＂ acute gouty arthritis （GA）, and to explore the mechanism. Methods： The 36 rats were divided into six groups as high-dose group, medium-dose group, low-dose group, western medicine group, model group and control group to establish a classical rat acute gouty arthritis model. Control group was injected into equal dose of normal saline. Perimeter （mean value） was measured in the same position of right ankle joint by binding method to evaluate the degrees of rat joint swelling. After 72 h, sample blood was taken from Abdominal aorta and the levels of IL-1 and TNF-α in blood serum were determined by ELISA, and numbers of leukocyte in adtevak were counted. Results： Rhizoma atractylodis-white tiger could significantly reduce the swelling of joints in rats with acute gouty arthritis, and lowerd the level of IL-1, TNF-α in blood serum and the numbers of leukocyte in adtevak. Among the groups, high-dose group was the most effective （P〈0.01）. Conclusion： Rhizoma atractylodis-white tiger can play a good role in resisting acute gouty arthritis by reducing IL-1 and TNF-α expression and inhabiting leukocyte hyperplasia.
