摘要
目的研究虫草多糖对腺嘌呤导致慢性肾衰模型大鼠转化生长因子-β1(TGF-β1)、单核细胞趋化蛋白-1(MCP-1)及信使核糖核酸(m RNA)表达的影响。方法选取SPF级清洁雄性SD大鼠60只,随机抽取20只作为正常对照组,其余40只利用腺嘌呤灌胃造成慢性肾衰竭模型,随机分为模型组20只、实验组20只。实验组给予虫草多糖160 mg/kg灌胃,对照组和模型组给予等量生理盐水灌胃,1次/d,共14 d。治疗结束后,采用生化分析仪分析血清肌酐(Scr)、尿素氮(BUN)含量,采用免疫组织化学法测定肾组织TGF-β1、MCP-1含量,采用RT-PCR法观察肾组织TGF-β1m RNA、MCP-1m RNA的表达情况。结果给药后与正常对照组比较,模型组、实验组大鼠的血清Scr、BUN值均明显较高,差异有统计学意义(P<0.05);实验组大鼠的血清Scr、BUN值明显低于正常对照组,差异有统计学意义(P<0.05)。治疗后正常对照组肾组织鲜有TGF-β1、MCP-1的表达,模型组肾组织的肾小管上皮细胞以及间质细胞中存在TGF-β1、MCP-1强表达,实验组肾组织存在TGF-β1、MCP-1表达,但弱于模型组;与正常对照组比较,模型组、实验组大鼠肾组织TGF-β1、MCP-1的OD值均明显较高,且模型组明显高于实验组,差异有统计学意义(P<0.05)。与正常对照组比较,模型组、实验组大鼠肾组织TGF-β1 m RNA表达量均明显较高,且实验组明显低于模型组,差异有统计学意义(P<0.05)。与正常对照组比较,模型组、实验组大鼠肾组织MCP-1 m RNA表达量均明显升高,且实验组明显低于模型组,差异有统计学意义(P<0.05)。结论虫草多糖能够减低腺嘌呤导致慢性肾衰模型大鼠的TGF-β1、MCP-1及m RNA表达,改善肾功能,对临床具有指导意义,值得临床推广。
Objective To explore the effect of Cordyceps Polysaccharide on the protein and m RNA expressions of TGF-β1and MCP-1 in rat model of chronic renal failure induced by adenine. Methods 20 rats from 60 SPF of clean male SD rats were randomly selected as control group. The remaining 40 rats were treated by adenine for chronic renal failure model construction. Rats of chronic renal failure model were randomly divided into model group and experimental group,20 rats in each group. Rats in the experimental group were given Cordyceps polysaccharide 160 mg/kg by gavage,rats in the control group and model group were given normal saline,once a day,14 d. After treatment,serum creatinine(Scr),blood urea nitrogen(BUN) content were analyzed by biochemical analyzer;Renal tissue TGF-β1,MCP-1 protein content was determined by immunohistochemical analysis. The expression of TGF-β1 m RNA and MCP-1 m RNA were determined by RT-PCR. Results After treatment,serum Scr and BUN in model group and experimental group were significantly higher than in the control group,the difference was statistically significant(P〈0.05);The levels of serum Scr and BUN in experimental group were significantly lower than that of control group,the difference was statistically significant(P〈0.05). After treatment,there was rarely expressed TGF-β1,MCP-1 in renal tissue of control group,but strong expression in renal tubular epithelial cell in renal tissue of model group and interstitial cells. There was also positive expressions of TGF-β1 and MCP-1 in renal tissue of the experimental group,but its expression levels was significantly lower than that of model group;The OD value of TGF-β1 and MCP-1 in renal tissue in the model group and the experimental group were significantly higher than in the control group,the difference was statistically significant(P〈0.05). The expression of TGF-β1 m RNA in the renal tissue in model group and experimental group were significantly higher than that in control group,and the experimental group was significantly lower than that in the model group,the difference was statistically significant(P〈0.05). The expression of MCP-1m RNA in renal tissue of model group and experimental group were significantly higher than in control group,and the experimental group was significantly lower than that in the model group,the difference was statistically significant(P〈0.05). Conclusion Cordyceps polysaccharide can significantly reduce the protein and m RNA expression of TGF-β1 and MCP-1 in chronic renal failure rats induced by adenine,and it also obviously improves the function of kidney. The results are worth of clinical application and promotion.
