刺山柑乙醇提取物乳膏对系统性硬皮病小鼠外周血循环内皮细胞等的影响 被引量:1

Effect of Capparis Spinosa ethanolic extract cream on circulating endothelial cells in systemic sclerosis mice

摘要 目的:观察刺山柑乙醇提取物乳膏对系统性硬皮病(SSc)小鼠外周血循环内皮细胞(CEC)数、血清一氧化氮(NO)及一氧化氮合酶(NOS)水平的影响。方法:BALB/c小鼠背部皮下注射盐酸博莱霉素诱导SSc模型,给予刺山柑乙醇提取物乳膏225,450,900 mg·kg^(-1)后,密度梯度离心法测定CEC数,硝酸还原酶法和ELISA法测定小鼠血清NO和NOS含量。结果:刺山柑乙醇提取物乳膏高剂量和阳性药青霉胺可降低小鼠CEC数,升高血清NO和NOS水平(P<0.01)。结论:刺山柑乙醇提取物乳膏对SSc血管内皮细胞损伤,NOS与NO释放减少有一定抑制作用。 OBJECTIVE To investigate the effects of capparis spinosa ethanolic extract cream on the number of circulating endothelial cells(CEC),nitrogen monoxidum(NO)and nitricoxide synthase(NOS)levels in bleomycin induced systemic sclerosis(SSc)mice.METHODS Mouse models with SSc were established by repeated local injections of bleomycin in BALB/c mouse back.After therapy by capparis spinosa ethanolic extract cream at 225,450 and 900 mg·kg^-1,the number of CEC was detected by density gradient centrifugation,the contents of NO and NOS in the serum were analyzed with nitric acid reductase methods and ELISA.RESULTS The number of CEC was markedly decreased,the levels of NO and NOS were markedly increased after administration of 900 mg·kg^-1 capparis spinosa ethanolic extract cream and 125 mg·kg^-1 penicillamine in bleomycin induced SSc mice(P〈0.01).CONCLUSION Capparis spinosa ethanolic extract cream is effective in ameliorating vascular endothelial cell injury and increasing NOS and NO levels in SSc mice.
