离子束重组菌DOB073的比较基因组学研究 被引量:4

摘要 为了深入认识低能离子注入驱动DOB基因组的突变机制,我们利用生物信息学方法,对离子束重组菌株DOB073进行了比较基因组学研究。结果表明,该菌株的基因组比原始菌株增加了12 883 bp,基因数目也比原始菌株增加了28个,但也缺失了原始菌株基因组中的13个基因。差异基因分析及其功能注释表明,离子束重组菌株DOB073基因组中新基因的功能主要与能量代谢、环境适应和DNA损伤修复相关,新基因主要分布于基因组的前部和后部;离子束重组菌株DOB073基因组中缺失的基因的功能主要与染色体分离蛋白和氯化物通道家族蛋白相关,主要分布于基因组的中部。基因组系统发育与进化的分析表明,离子束重组菌株DOB073与原始菌株亲缘关系最近。差异基因的COG功能富集分析表明,离子束重组菌DOB073独有的基因功能明显富集于蛋白质翻译、核糖体结构和发育、细胞壁/细胞膜/胞外层发育、防护机制,而碳水化合物转运与代谢则明显存在于原始菌株的独有基因中。本研究为低能离子注入驱动微生物的全基因组突变与重组提供了一定的分子证据,也为低能离子注入驱动新基因的从头起源研究提供了一定的理论和实践依据。 In order to deeply understand the mutation mechanism of DOB genome driven by low-energy ion beam implantation,we carried out the comparative genomics study of the ion beam recombinant bacteria DOB073 by using bioinformatics methods.The results showed that the genome of DOB073 was 12 883 bp higher than that of the original strain,and the number of genes was 28 more than the original strain,but it also lacked 13 genes which was existed in the original strain genome.The differential gene analysis and functional annotation suggested that the functions of new genes in recombinant bacterial strain DOB073 were mainly related to energy metabolism,environmental adaptation and DNA repair,and these novel genes were principally distributed in the front and back of DOB073 genome;the functions of deleted genes in recombinant bacterial strain DOB073 were mostly relevant to chromosome segregation protein and chloride channel family protein,and these genes were located in the middle of the genome.Phylogenetic and phylogenetic analysis of genomes indicated that DOB073 genome had the closest relationship with the original strain.The COG functional enrichment analysis of differential gene implied that the unique gene functions of DOB073 were enriched in protein translation,ribosome structure and development,cell wall/cell membrane/cell outer layer development,and protection mechanism,while the carbohydrate transport and metabolism was obviously existed in the unique genes of the original strain.Our research provided some relative molecular evidences for inducing the whole genome mutations and recombination driven by low-energy ion beam implantation and supplied some theoretical and practical bases for the de novo origination research of novel genes driven by low-energy ion beam implantation.
