摘要
目的研究芒鲶胶原组织再生膜(Paugusiushamiltoa collagen tissue regeneration membrane,简称PCM)的制备、物理性质、生物学特性以及引导骨组织再生的特性。方法利用芒鲶鱼皮胶原蛋白为原料,采用脱色、脱脂、脱细胞和热交联方法制备组织再生膜。通过电镜扫描(SEM)、吸水率、孔隙率和力学强度评价其理化特性。通过兔颅骨骨缺损模型,评价PCM引导骨组织再生的效果。结果 SEM扫描结果显示,PCM呈双层膜结构,一侧为网状多孔结构,另一侧光滑致密;其吸水率约为自身质量的4倍,孔隙率为78%,平均拉伸应力最大值为18.82MPa;皮内刺激反应显示PCM未造成红斑、焦痂形成和水肿,对皮肤无刺激性;以瑞士Bio-Gide膜作为阳性对照组,在4周和8周后,通过显微CT、苏木精-伊红染色(HE染色),兔颅骨骨缺损修复动物实验表明,PCM能屏障纤维结缔组织,诱导成骨细胞生长,具有引导骨再生的作用。结论 PCM具有优良的生物医用材料特性,具有引导骨再生的作用,是优良的引导骨再生修复材料。
Objective Tissue regenerating Paugusiushamiltoa collagen membrane was prepared and the physical and biological characteristics were evaluated for bone tissue regeneration. Methods The bone tissue regenerating membrane was derived from Paugusiushamiltoa and prepared by decolorization, de- greasing, decellularization and cross-linking. The physical and biological properties were evaluated with microstructure,porosity, water absorption, mechanical and degradation properties. The effects of bone repair were evaluated by calvarial defect models of New Zealand rabbit. Results The bilayer structure including a dense layer and a loose layer of the surface morphology of PCM was revealed by SEM. The porosity rate, water absorption rate and average tensile stress were 78%, 4 times than itself and 18.82 MPa respectively. Intradermic reaction test showed that PCM could no skin irritation. The repairing effects were presented by Micro-CT and the image analysis of the hematoxylin-eosin staining (HE) of calvarial defect models with PCM, Bio-Gide membrane as positive control after 4 and 8 weeks post-op- erat^on. The results revealed that PCM could restrict the growth of fibrous connective tissue in the rep- arative process and promote new bone regeneration. Conclusion PCM featured excellent biomedical ma- terial property and could guide new bone accumulation. It was recommended to apply Paugusiushamil- toa collagen membrane as bone regeneration substitute.
