黄连解毒汤对胰岛素抵抗大鼠炎症因子和氧化应激水平的影响 被引量:21

Effects of Huanglian Jiedu Tang (黄连解毒汤) on Inflammatory Factors and Oxidative Stress of Insulin Resistance Rats

摘要 目的:观察黄连解毒汤对胰岛素抵抗大鼠IL-6、TNF-α和SOD、MDA的水平的调控作用,探讨其作用途径。方法:雄性SPF级Wistar大鼠60只,随机分为6组,正常组,模型组,黄连解毒汤高、中、低剂量组,二甲双胍组。高脂高糖高盐喂饲大鼠8周复制胰岛素抵抗模型,造模成功后连续给药2周,末次给药后后禁食12 h,采血检测空腹胰岛素(Fasting insulin,FINS),胰岛素敏感型指数(Insulin sensitivity index,ISI),空腹血糖(Fasting plasma glucose,FPG)及大鼠血清SOD、MDA、IL-6和TNF-α的水平。结果:与模型组比较,黄连解毒汤各组血清FPG,FINS水平均明显下降(P<0.05或P<0.01),ISI值上升(P<0.05或P<0.01);SOD水平显著提高,MDA含量和IL-6、TNF-α水平明显下降(P<0.05或P<0.01),以高剂量组效果最为显著。结论:黄连解毒汤可显著改善IR模型大鼠的胰岛素抵抗,疗效呈剂量依赖趋势,其作用机制与调节炎症因子、氧化应激水平有关。 Objective: To observe the effects of Huanglian Jiedu Tang on inflammatory factors ( IL - 6, TNF -α) and oxidative stress indexes (SOD,MDA) in insulin resistance rats and explore its mechanism. Methods: 60 male Wistar rats were randomly divided into normal group, model group, Huanglian Jiedu Tang group high dose group, middle dose gorup, low dose group and metformin positive group. Rats were fed high - glucose, high - fat and high - salt diet for 8 weeks to establish insulin resistance model and then were given the corresponding medicine for 2 weeks, after the last administration, followed by detecting the level of FPG, FINS, and 1SI, and the levels of serum SOD, MDA, IL -6 and TNF -α were measured after fasting for 12 hours. Results: Compared with the model group, serum FPG and FINS were significantly decreased in each Huanglian Jiedu Tang group (P 〈0. 05 or P 〈0. 01 ), ISI was significantly increased (P 〈0. 05 or P 〈0. 01 ), serum SOD activity was raised, while levels of MDA, IL - 6 and TNF - α were lowered ( P 〈 0. 05 or P 〈 0. 01 ) , and were in a dose - dependent manner. The effects of the Huanglian Jiedu Tang high dose group was the most. Conclusion: Huan- glian Jiedu Tang can significantly improve insulin resistance in model rats, and its effect is related to the dose, its mechanism might be correlated with the regulation of inflammatory factors and oxidative stress.
